Objective Pyrroline-5-carboxylate reductase 1(PYCR1)is an enzyme involved in cell metabolism,which has been shown to be up-regulated in cancers.However,the functions of PYCR1 in prostate cancers(PCa)are still largely unknown.Methods(1)Knocked down PYCR1 in PCa cell lines(DU145,PC-3)via lentivirus-mediated gene delivery and analyzed its biological function.(2)PYCR1 specific si RNA vector was constructed then infected the prostate cancer cells with recombinant lentiviruses,meanwhile control group and sh PYCR1(S1)and sh PYCR1(S2)transfected group were set up.RT-PCR and Western blotting were used to detect the expression of the PYCR1 m RNA and protein in Prostate cancer cells.(3)MTT assay,PI assay and Annexin V-APC/7-AAD assay were used to detect the proliferation,cell cycle and the apoptosis of the prostate cancer cells.Results The expression of PYCR1 at m RNA and protein levels was reduced significantly(P<0.01),suggesting that the PYCR1-silencing cell line sh PYCR1 was successfully established.The cell proliferation and its clone formation of sh PYCR1(S1)and sh PYCR1(S2)group was suppressed when compared with control group(P<0.01),which also caused the decrease of cell cycle regulatory proteins and the increase of cell apoptotic-related proteins(P<0.01).Conclusion PYCR1 is high expressed in the PCa cell.Knockdown the PYCR1 gene in PCa cell suppressed the cell proliferation and colony formation,suggesting that PYCR1 plays a role in the prostate cancer development and may be a potential tumor marker.
