Objective: 1)To isolation,purification and analyze basic chemical structure of polysaccharides from Fomes Officinals(FOPS)of uyghur medicine;2)To determine the in vitro anti-oxidant activity and protective effect on oxidative DNA damage of polysaccharides from Fomes Officinals.Method: 1)The crude polysaccharides from Fomes Officinals were isolated and purified by DEAE-52 cellulose ion-exchange chromatography,sepharose and sephadex gel chromatography.2)The purity of polysaccharides was evaluated by gel filtration method,UV method and SDS-PAGE analysis.Molecular weight of polysaccharides was evaluated by gel filtration method.3)The spectral analysis of GC,GC-MS,IR,NMR and chemical methods of methylation analysis,partial acid hyrdrolysis,periodate oxidation and Smith degradation were used to analyze basic chemical structure the polysaccharides with high content.4)Protective effect on oxidative DNA damage,savenging on DPPH,hydroxyl radical,superoxide anion and determination of reducing power,were used to evaluate antioxidant activity of FOPS.Results : 1)Trough isolation and purification methods two homogeneous polysaccharides of FOPS-a and FOPS-b from Fomes Officinals were obtained.2)Average molecular weight of FOPS-a was 199 kDa,and it was composed of glucose,xylose,arabinose,mannose and galactose,the ratio is 0.012?1?1.90?2.38?6.25.and its main chain was formed by a mixing linkage of 1,3,6-galactose,1-glucose,and side chain were attached at 1,2-xylcose,1-mannose,1,2,6-mannose,1,3-galactose;Average molecular weight of FOPS-b was 87 kDa,and it was composed of arabinose,mannose,glucose,xylose and galactose,and the ratio is 1?2.01?0.93?1.24?4.3.its main chain was formed by a mixing linkage of 1,4-galactose,1,6-glucose,1,4-mannose and side chain were attached at 1,6-mannose?1,3-glucose,1,3,6-galactose and 1,4-galactose.4)The antioxidant activity analysis results indicated that crude polysaccharises and fractions FOPS-a and FOPS-b from Fomes Officinalis have reducing capacity,hydroxyl free radical scavenging ability,DPPH free radical scavenging and superoxide anion scavenging ability,UV method and SDS-PAGE analysis results show that,protective effect on oxidative DNA damage of polysaccharides from Fomes Officinalis had an effect with various levels of oxidative DNA damage induced by hydroxyl free radical·OH.Conclusion: 1)It has been obtained two polysaccharides fractions of FOPS-a and FOPS-b were isolated and purified from Fomes Officinalis.Structural elucidation show that FOPS-a and FOPS-b are complicated heteropolysaccharides with multi-branch.And monosaccharide component of two polysaccharides had differences each link between sugar sequence and glycoside type 2)Antioxidant activities analysis show that polysaccharides from Fomes Officinalis had certain in-vitro antioxidant activity,and it was providing the basis for further research on relationship of structure and activity,and comprehensive utilization of the polysaccharides.
