In this study,we isolated polysaccharides from Polygala tenuifolia Willd and Alhagi pseudalhagi Desv.Through a series of purification steps,eight polysaccharides were isolated and purified from P.tenuifolia and A.pseudalhagi.Firstly,eight raw saccharides were obtained by water/NaOH extraction and ethanol precipitation from P.tenuifolia and A.pseudalhagi,which noted as PT50,PT70,PT90,PTPB,AP50,AP70,AP90 and APPB.Subsequently,pure polysaccharides(labeled as PTP70-2,APP50-1-1,APP50-1-2,APP50-2,APP70-1,APP70-2,APP70-3-1 and APP70-3-2)were isolated and purified from raw saccharides PT70,AP50 and AP70 via DEAE-Cellulose-52 and Sephacryl S-100.The results of tiny difference in specific optical rotation of two precipitates and high performance gel permeation chromatography(HPGPC)suggesteded that PTP70-2,APP50-1-1,APP50-1-2,APP50-2,APP70-1,APP70-2,APP70-3-1 and APP70-3-2 were purificatory.The relative molecular weight of PTP70-2,APP50-1-1,APP50-1-2,APP50-2,APP70-1,APP70-2,APP70-3-1 and APP70-3-2 were found to be 65.13 kDa,27.02 kDa,11.43 kDa,6.01 k Da,3.42 k Da,3.94 kDa,9.06 kDa and 9.37 kDa,respectively.The analysis of monosaccharide constituents,determination of infrared spectroscopy(IR),a few chemical reaction,gas chromatography-mass spectrometry(GC-MS)and nuclear magnetic resonance(NMR)spectrums were carried out to characterize the primary structures of eight polysaccharides.The congo-red assay showed eight homogeneous polysaccharides have not triple helical structure.The scanning electron microscope(SEM)assay showed the microstructure and morphological characteristics of eight polysaccharides were distinctive entirely.Besides,the structure of eight polysaccharides were obtained for the first time.
