Objective: To investigate the application value of targeted ultrasound constrast agent(MBt)with Anti matrix metalloproteinase 2(MMP2)monoclonal antibody in vasculogenic mimicry(VM)in ovarian transplantation tumor of nude mice.Methods: MBt was prepared by biotin-avidin method.The conjugation was demonstrated by immunofluorescence;The tumor transplanted subcutaneously model of SKOV3 in nude mice was established and underwent ultrasound contrast examination with MBt or control microbubles(MBc).Relative parameters of time-intensity curves(TIC)were analyzed and MMP2 expressions and vasculogenic mimicry of ovarian cancers were tested by immunohistochemistry.Results: MBt carrying with MMP2 was prepared successfully;TIC showed that MBt group TTP23.57(7.58,29.8)had earlier the time to maximum echoenicity(P<0.01)than MBc group TTP 33.37(25.35,38.38),and MBt group S545.00(466.50,609.50)had longer lasting time(P<0.01))than MBc group S 342.00(237.00,540.00),there was no significant difference in peak intensity between two groups(P>0.05),The second peak value was observed in peak intensity of MBt group;High MMP2 expression was discovered by immunohistochemistry in vasculogenic mimicry of tumor tissues.Conclusions: MBt can specifically bind to vasculogenic mimicry with specific expression of MMP2 in vivo,and be well evaluated vasculogenic mimicry of ovarian canaer.
