Objective:Subcutaneous transplantation model of ovarian cancer SKOV3 was established in nude mice and divided into different stages.To study the imaging features of vasculogenic mimicry?VM?with matrix mimicrymetalloproteinase-2?MMP-2?targeted ultrasound contrast agent in different stages of ovarian cancer transplantation.Methods:Fifty nude mice bearing ovarian cancer xenografts were randomly divided into 5 groups,14 days,21 days,28 days,35 days and 42 days after transplantation,a group of them were selected to study the characteristics of normal and target ultrasound contrast,then all mice were sacrificed,and the tumor masses were staged by pathology.To observe the expression of VM and MMP2 in different stages of tumor transplantation and to study the correlation with the peak intensity?PI?and duration?TTD?of the target contrast parameters.Results:There was statistical significance in different stages of ovarian cancer xenograft VM density difference?P<0.05?,the II stage expressed highest;immunohistochemistry confirmed the high expression of MMP2 in peripheral and internal of VM,and positively correlated with I and II(rI=0.647,rII=0.728,P<0.01),no correlation with III the density of VM?P>0.05?.The PI and TTD of stage I and II target contrast were higher than those of ordinary contrast,and the difference was statistically significant,and there was a positive correlation with VM density?P<0.05?.Conclusions:MMP-2 targeted ultrasound imaging can evaluate the early VM imaging of ovarian cancer and provide new ideas and new basis for early diagnosis of ovarian cancer.
