| Objective : According to the clinical efficacy of Chinese medicine preparation XueNiao capsule,the curative effect of hematuria caused by acute pyelonephritis and renal contusion is obvious,but the basic research is weak.The subject is screened by antibacterial,diuretic and sIgA content.And on the whole level to explore the effect of XueNiao capsule in the treatment of acute pyelonephritis and mechanism for the clinical application of XueNiao capsule to provide the basis for the XueNiao capsule prescription and the preparation process rationality and secondary development research to provide the basis.Methods : The antibacterial test in mice,the diuretic experiment in rats and the detection of sIgA content in the urethra were carried out to screen the four parts of the XueNiao capsule prepared by the system solvent method.The basal basis of the XueNiao capsule was established.The rat model of acute pyelonephritis,with XueNiao capsule and its four parts of the administration,the overall level of detection and acute pyelonephritis caused by renal function,inflammation and immune-related changes in indicators,explicitthe effect of XueNiao capsule in the treatment of acute pyelonephritis.The metabolic pathways of acute pyelonephritis were identified by nuclear magnetic resonance spectroscopy(1H NMR)and to evaluate the mechanism of XueNiao capsule in the treatment of acute pyelonephritis from the whole level.Results:The results of antimicrobial experiments showed that the leaching rate of petroleum ether and ethyl acetate was 10% and 10%,respectively.The results of diuretic test showed that the urine output of petroleum ether and ethyl acetate was significantly higher than that of other(P <0.01).The level of sIgA in urethra was significantly higher than that in other administration groups(P <0.01),while the level of sIgA in urethral capsule was significantly higher than that in other administration groups(P <0.01)Higher than the other administration groups(P <0.01).Acute urinary pyelonephritis model showed that XueNiao capsules could reduce the contents of urine and kidney bacteria(P <0.01)compared with the model group.In the detection of blood and CD4 and CD8,the blood samples of white blood cells and lymphocytes(P <0.01),while XueNiao capsules and petroleum ether sites were able to significantly regulate CD4 / CD8,which tends to be blank(P <0.01);XueNiao capsules and other parts(IL-1?,IL-1?,IL-6,IL-10 and CXCL-2)were significantly lower than those in the control group(P <0.01),and the levels of IL-(P<0.01).The renal tissue index of XueNiao capsules significantly reduced the renal index(P<0.01),while the petroleum ether and ethyl acetate sites of XueNiao capsules significantly increased the ratio of left and right kidney(P <0.01,P < 0.05).The renal histopathological findings of the XueNiao capsule group were found to be substantially complete,and the degree of inflammatory cell infiltration was significantly less than that of the other administration groups.Metabolomics studies showed that the endogenous metabolites of the ligation of the kidneys in the rats of each administration group were identified as 37 compounds,including amino acids,organic acids and carbohydrates.The partial least squares discriminant analysis showed that 23 differential metabolites,the database detection and access to literature,obtained XueNiao capsule in the treatment of acute pyelonephritis related metabolic pathways have energy metabolism,lipid metabolism,branched-chain amino acid metabolism and valine,leucine,isoleucine Biosynthetic pathway;glycine,serine,threonine metabolic pathway;alanine,aspartic acid,glutamic acid metabolic pathway and methane metabolic pathway etc.Conclusion:Studies have shown that XueNiao capsules from the overall regulation of treatment of acute pyelonephritis is through the various parts of the common play,in which the petroleum ether and ethyl acetate parts have a clear antibacterial and long-acting diuretic effect,XueNiao capsule quick effect of diuretic was significantly stronger than the rest The antimicrobial effect of XueNiao capsules in each administration group and acute pyelonephritis model may be related to the increase of sIgA content and the regulation of immune function,and its anti-inflammatory effect may be achieved by down-regulating the level of inflammatory cytokines,Metabolomics research from the overall level of evaluation of XueNiao capsule in the treatment of acute pyelonephritis affected by the various parts of the metabolic pathway,basically clear its mechanism of action.Therefore,XueNiao capsule recipe and extraction preparation process is reasonable,to ensure the full play of the efficacy. |
