To Observe The Effect Of Decoction That Replenish Spleen&Qi,Move Bowels On The Expression Of C-kit Protein Of Rats With Slow Transit Constipation

1 ObjectiveTo observe the effect of Decoction that replenish spleen& qi,move bowels on the expression of c-kit protein of rats with slow transit constipation and to explore the possible action mechanism of Decoction that replenish spleen& qi,move bowels on slow transit constipation.2 Methodsforty adult male rats were randomly divided into four groups: blank group,model group,Mosapride group,group of Decoction that replenish spleen& qi,move bowels,ten rats in each group.Build model rats group with slow transit constipation by hypodermic injection of morphine at the dose of 2.5 mg / kg daily for forty-five days(except the blank group),verify whether the models is feasible by intestinal propelling.The control group: no model,conventional feeding;model group: feeding saline by gavage 2ml/bid;Mosapride group:feeding 5mg of grained Mosapride each time by gavage,two times a day;Decoction group: feeding the replenish spleen& qi,move bowels Decoction that contains crude drug 2ml/100g/bid by gavage.Each group has ten rats,which are allowed free eating and drinking during the period of gavage.The general condition of the rats in each group was observed every day,and the mental state,food,activity and stool were recorded.Model group,mosapride group and Decoction group conduct the gavage from one day after the finishing of the model,keep medication for four weeks than stopping one week,feeding the common feed to be processed.Checking the rate of bowel movement and the pathological features of colonic mucosa of rats in each group.The expression of c-kit protein was detected by Western-blot.3 Results(1)Compared with the blank control group,the stool of the model group was obviously dry,the granule number of defecation was decreased,the rate of carbon propulsion of the small intestine descended,the normal structure of colon tissue was destroyed,the expression of C-kit protein decreased significantly(P <0.01).(2)Compared with the model group,the fecal characters of the rats in Mosapride group and Decoction group were improved,especially in the latter group the granule of rats gradually enlarger than before,shaped in ball to column,the rate of carbon propulsion of the small intestine increased,oedema of colonic mucosa obviously relieved,the expression of C-kit protein increased significantly(P <0.01).(3)Compared with Mosapride group,the expression of C-kit protein in Decoction group increased significantly(P <0.05).4 ConclusionThe incidence of slow transit constipation is related to the down-regulation of C-kit expression in colonic mucosa,Decoction that replenish spleen& qi,move bowels can improve the propulsive rate of small intestine and relive the slow transit constipation symptoms and mucosal chronic inflammation,improve the number and distribution of Cajal interstitial cells by up-regulating the expression of C-kit to cure STC.
