Poria cocos, also named Fuling, is the dried sclerotia of Poria cocos (Schw.) Wolf, which is a kind of Fungi belonging to PolyPoraceae. the surface layer of Poria cocos (Fu-Ling-Pi in Chinese) is the epidermis of the sclerotia. Poria cocos often was used as an diuretic, sedative and tonic herb in traditional Chinese medicine. It contains triterpenoids, polysaccharides, Fatty acids and amino acids. Some of these compounds showed anti-emetic, anti-inflammatory, anti-tumor, and exhibit an activity of immune function. For many decades, the chemical composition of Poria cocos already have a lot of researchs, Studies have shown that it has a significant biological activity and prove ttriterpenoids are the characteristic ingredients of Poria cocos, is also the main medicinal ingredient, Therefore this Studiy has important significance. To further clarify the biological active ingredients of Poria cocos, And to provide the material and the theoretical basis for quality control, clinical applications, and further development of the resource, So in this thesis, chemical composition of triterpenoid acids were prepared.In this paper, twelve triterpenoid acids compounds were isolated from the surface layer of Poria cocos by a variety of means of separation, including silica gel column chromato-graphy, recrystallization and the high pressure preparative liquid chromatography. By purity testing, the purity of ten compounds more than 90%, and of which six compounds’purity more than 98%. Their structures were identified as poricoic acid AM(1), poricoic acid A (2), poricoic acid B(3),3-o-acetyl-16a-hydroxytrametenolic acid(5), dehydropachymic acid(6), dehydrotramet -enonic acid(7), dehydroeburiconic acid(8), dehydrotrametenolic acid(9), eburicoic acid (10),3-o-formyl-dehydrotrametenolic acid(11) and 3-o-formyl-eburicoic acid(12). And the two new compounds 11 and 12 are artifact produced during isolation by validation experiments.
