Objective:1.Through the research on the herbal medicine to have a comprehend of the medicinal history of Polygonum cuspidatum.2.Through the investigation of the resources in Shitai to have a grasp of the distribution and development conditions of Polygonum cuspidatum.3.Through the detection and correlation analysis of 20 inorganic elements in Polygonum cuspidatum and its Rhizosphere soil, study the enrichment of 20 inorganic elements from its rhizosphere soil. The conclusion can provide a reference for the development of selenium-rich herbal industry in Shitai.4.Through the analysis of inorganic elements and organic components in Polygonum cuspidatum, provide a scientific basis for the establishment of quality standard system of Polygonum cuspidatum.Methods:1.Through the field investigation and market access, master the resource distribution and utilization of Polygonum cuspidatum.2.The contents of 20 inorganic elements in Polygonum cuspidatum and its rhizosphere soil were determined by inductively coupled plasma mass spectrometry(ICP-MS),and the correlation analysis was carried out by using statistical software.3.The contents of emodin, polydatin and resveratrol in the cortex, phloem, xylem and myeloid of Polygonum cuspidatum were determined by high performance liquid chromatography.Results:1.Polygonum cuspidatum has a long history of medicine used, and have a higher development value.2.The distribution of Polygonum cuspidatum was mainly concentrated in the central and western regions of Shitai. The local Polygonum cuspidatum were all derived from wild mining. The annual acquisition were about 40 tons.3.The soil content of Se in Shitai reached the selenium-enriched level, among which the highest content of soil is Yuantou village 5.06 mg / kg.The contents of As and Cd of the soil in several towns exceeded the limit.4.The level of the 20 elements in Polygonum cuspidatum is Ca> K> Mg> Ba> Al> Fe>Na> Sr> Mn> Zn> Cr> Cu> Ti> Mo> Pb> Se> Co> As> Cd> Hg;The results showed that the content of Se in all 7 areas were all reached selenium-enriched level except Renli, and the range is 0.26-1.9 mg · kg-1.The content of heavy metals in all the samples was within the standard level. The principal component analysis show that Ba,Se,Cr,Na,Zn,Al and Ca are the characteristic elements of Polygonum cuspidatum. The correlation analysis show that Al was significantly correlated with As,Ba,Fe,Mo,Pb,Se,Ti,and Ca was significantly correlated with Mg,Na and Sr,and Se was significantly correlated with Ba and Mo.5.Most of the elements were positively correlated with their rhizosphere soil elements, and the correlation with Se,Mo and Ba was significant. Most of the elements were negatively correlated with diameters. The negative correlation with Mg and Al was significant. There were no significant differences in the content of inorganic elements between the different harvesting months. The contents of 20 kinds of inorganic elements in leaves, stems and rhizomes were different, but they had similar enrichment laws. The difference between root and rhizomes is small in the content of inorganic elements. The difference in Skin, phloem, xylem and pith of the 20 kinds of inorganic elements were significant: the highest were pith and skin, followed by phloem and xylem. There are similar laws in different areas of Polygonum cuspidatum The content of Se,Mo and Ba in Shitai County were significantly Higher than other areas. There was no obvious rule between the male and female plants.6.The content of emodin in the seven townships was 0.395%-1.064%,the content ofpolydatin was between 1.374% and 6.129%,and the content of resveratrol was between 0.133% and 0.965%.The content of the skin, phloem, xylem and the pith of the three organic ingredients were obvious difference. The enrichment of inorganic elements has similar characteristics with the elements, for the pith and skin has the highest content, followed by phloem and xylem.Emodin and resveratrol were negatively correlated with the diameter, and the correlation was P <0.05.The polydatin was positively correlated with the diameter. The contents of Na,Mg,Sr,Ca and K were significantly correlated with emodin.Al was positively correlated with polydatin, and Na,Mg,K,Ca,Ti and Fe were significantly correlated with resveratrol.Conclusions:1.Through field survey, know well with the distribution and reserves of Polygonum cuspidatum, understand the level of development and utilization. It can lay the foundation for the follow-up research work.2.ICP-MS was used to determine the selenium level of local herbs firstly, and also has a correlation analysis to the content of inorganic elements of soil and herbs. The status of soil heavy metal pollution was also investigated. The results can provide theoretical basis for the development of Se-riched medicine industry.3.By analyzing the content of different tissues and different diameters of inorganic elements, reveal the accumulation characteristics of inorganic elements in Polygonum cuspidatum, which can provide reference for the specification of the classification of Polygonum cuspidatum.According to the content of different harvest period, male and female plants and different origin of Polygonum cuspidatum, provide a basis for reasonable development the of resources for Polygonum cuspidatum.4.Through the analysis of three organic components of Polygonum cuspidatum in several villages and towns in Shitai County, the quality of the local herbs was investigated. In addition to a few samples of emodin content is not up to the outside, the rest of the organic ingredients are in line with the "Chinese Pharmacopoeia" standard. The results showed that the overall quality is better.
