Objective: Background: with the development of social economy and living standards continue to improve and change the way of life,a higher intake of calories daily catering,and sedentary lifestyles are becoming more and more popular,resulting in excess energy and fat accumulation.Obesity refers to excessive accumulation of fat in the body and(or)abnormal distribution,weight gain,genetic factors,environmental factors and other factors caused by chronic metabolic diseases.Overweight and obesity are endemic in the world,and have become one of the serious public health crises.As one of the main components of metabolic syndrome,obesity is closely related to many diseases such as type 2 diabetes mellitus,dyslipidemia,hypertension,coronary heart disease,stroke,cancer and so on.Excessive accumulation of fat in the internal organs,resulting in visceral organ fat like change,the most common is the fatty degeneration of the liver,known as fatty liver.Fatty degeneration of liver cells in the blood coagulation factor and albumin synthesis and degradation of toxins and bilirubin cycle life important metabolic process can not be carried out smoothly,and thus lead to the occurrence and development of a series of metabolic diseases.So we determine to evaluate the effects of treatment with sitagliptin on the oxidative stress and metabolism of glucose and lipid in diet-induced obese complicated with non-alcoholic fatty liver rats.Methods: All SD rats were randomly separated into 2 groups: the normal food group(ND)and the high fat food group.The rats in the high fat food group were fed with high energy and high fat food 16 weeks to establish obesity with non-alcoholic fatty livermodels,then the model rats were randomly divided into the obesity group(OB),the Sitagliptin group(ST).Every group includes 8 rats in above three groups.After 16 weeks,the rats in the ST group were treated with Sitagliptin for 6 weeks,the rats in theND and OB group were treated with equal normal saline for 6 weeks.Measuring Body weight(BW),fasting blood glucose(FPG),fasting insulin(FINS),triglyceride(TG),serum SOD,MDA,GSH-Px respectively before and after the 6 weeks of treatment and GSH-Px,SOD,MDA,TG in 10% liver homogenates after sacrifice of the rats,and calculate HOMA-IR,oil red O staining were used to assess pathological changes in livers.Results:(1)Before treatment,all thediameters in the OB and ST group were of no statistical significance,compared with OB and ST group,the diameters in the ND group: the level of FPG,FINS,TG,HOMA-IR,MDA were lower(P<0.05),the level of SOD and GSH-Px were higher(P<0.05).(2)After treatment,compared ST and ND group,the level of TG and MDA in the liver homogenates of ST group were higher than ND group(P<0.05),the level of FPG,FINS,serum TG,HOMA-IR,serum MDA,SOD GSH-Pxin both serum and liver homogenates of ND group and ST group were of no statistical significance.The rats liver fatty degeneration in the ST group were more severe than those in the ND group,compared with OB group,the levels of FPG,TG,FINS,HOMA-IR and serum level of MDA and TG in ST group decreased while the level of SOD and GSH-Px in the serum and liver increased with a statistical significance(P<0.05).Meanwhile,the rats liver fatty degeneration in the ST group were less severe than those in the OB group.Conclusion: Sitagliptin intervention can improve glucose and lipid metabolism,reduce liver steatosis maybe associated with alleviating oxidative stress.
