The Effects Of Zedoary Turmeric Oil On Thyroid Anaplastic Carcinoma Doxorubicin Resistant Strains (HTh74Rdox)

Objectives:To explore the influence of Zedoary Turmeric Oil in proliferation and drug resistance of HTh74Rdox cells and how did these produce.Methods:HTh74Rdox cells were to be treated by different concentrations(0?60?90?120mg/L)of Zedoary Turmeric Oil,in an effort to observe the morphological change and apoptosis situation of HTh74Rdox cells;MTT was used to test inhibiting effect of Zedoary Turmeric Oil on the proliferation of HTh74Rdox cells;The cell cycle change and cell apoptosis rate were to be further detected by flow cytometry;Western Blot was used to measure the expressions of Bcl-2 and Bax;And finally,with the help of qPCR,the mRNA expressions of MDR1 and ABCG2 were to be detected.Results:With different concentrations of Zedoary Turmeric Oil(0,60,90,120 mg/L)processing after 24 hours,HTh74Rdox cells were observed under inverted microscope cell morphology,can we see that,since Zedoary Turmeric Oil concentration is 60 mg/L,some cells began to be divorced from the petri dish;With the increasing of concentration of Zedoary Turmeric Oil,cell density coefficient and adherent cells decreased,at the same time,cell morphology became into pieces.When Zedoary Turmeric Oil concentration increased to 120 mg/L,almost all cells were divorced from the petri dishes,cell shape completely became into irregular shape and cell death together led to cell sap turbidity.MTT showed that Zedoary Turmeric Oil played an significant role in inhibiting the growth of HTh74Rdox cells.Flow cytomety showed that Zedoary Turmeric Oil induced cell apoptosis,especially the early apoptosis(P<0.01).Western Blot showed that Zedoary Turmeric Oil significantly increased the percentage of Bax/Bcl-2(P<0.05).qPCR showed that Zedoary Turmeric Oil reduced the mRNA expressions of MDR1 and ABCG2.With single medicine Zedoary Turmeric Oil,single medicine Adriamycin or combination of Zedoary Turmeric Oil and Adriamycin processing after 24 hours,comparison 2.0 mg/L Adriamycin + 90 ml/L Zedoary Turmeric Oil group with 2.0 mg/L Adriamycin + 0 ml/L Zedoary Turmeric Oil group,were statistically difference(P<0.05),which prompted that combination of Zedoary Turmeric Oil and Adriamycin enhanced Adriamycin cytotoxic effect.Comparison 1.0 mg/L Adriamycin + 45 ml/L Zedoary Turmeric Oil group with 1.5 mg/L Adriamycin + 45 mg/L Zedoary turmeric oil group were not statistically significant(P>0.05),which prompted that combination of Zedoary Turmeric Oil and Adriamycin reduced the average dose for Adriamycin.Conclusions:Zedoary Turmeric Oil can inhibit the growth of HTh74Rdox cells and improve its drug resistance significantly.And the possible mechanisms for Zedoary Turmeric Oil are related to the increase of Bax/Bcl-2 ratio and the reduction of MDR1 and ABCG2 mRNA expressions respectively.
