Objective:IBS is abdominal pain abdominal pain or discomfort,shit with characters and bowel habit change characterized by chronic or recurrent gastrointestinal disorders bowel disease.Compared with adults with levels of diseases such as diabetes,congestive heart failure,cirrhosis of the liver and renal insufficiency,IBS greatly influenced the quality of people’s lives.It is reported that in 2013 the incidence of IBS in Europe and North America is close to 20%.The incidence of domestic for 10～15%.The prevalence of Asian countries are gradually growth,has gradually close to the prevalence of European and American countries.In IBS,change the IBS is the most common type witness,to change is the basic pathogenesis.Clinical treatment on liver spleen for therapeutic principle.The treatment mechanism may be related to lower intestinal sensitivity,increase of intestinal pressure threshold,regulating gastrointestinal hormone secretion,interactive features,regulation of the immune system,enhance immunity.At present the study of pathogenesis is not fully clear.Through clinical observation,Ann intestinal antidiarrheal party to change the type of IBS-D had a very good curative effect,Hope that through this study can understand Ann intestinal antidiarrheal party to change the type of IBS-D rats serum and brain gut hormone VIP on intestinal mucosa is influential,5-HT.Methods:(1)molding:50 SPF SD rats,senna colon perfusion and chronic stress method to build model bound + cold water stimulation.Evaluate the results of building after building.(2)grouping:SD rats according to random number table method were randomly divided into blank group,model group,maleic acid trimebutine group,intestinal antidiarrheal low and high dose group.(3)Drug delivery:The normal group and model group with normal saline lavage.Maleic acid trimebutine maleate group to trimebutine 0.03 g/ml lavage,volume 20 ml/kg,Ann intestinal antidiarrheal the high and low dose group,respectively with 1.5 g/ml,0.75 g/ml concentration of traditional Chinese medicine(TCM)solution to fill the stomach,2 times a day,for two weeks.(4)measurements:a.Building phase changes in the rate of loose stools in rats.b.Balloon expansion experiment testing rats colon pain sensitivity.c.Using enzyme-linked immunoassay detection of serum and VIP in colon tissue,5-HTlevels.Results:Rat made module loose stools rate is significantly higher than the first day,3 days 7 days reduced.Compared with the blank group,the rectum expands,model group raised his abdomen and back up the stress threshold significantly reduced(P<0.01),compared with model group,maleic acid trimebutine,Ann intestinal antidiarrheal high dose groups abdominal significantly increased lift up and back up the threshold pressure(P).Model group serum and colon tissue VIP,5-HT content than the blank group increased significantly,maleic acid trimebutine group of western medicine and traditional Chinese medicine(TCM)Ann intestinal antidiarrheal party high dose group can make the model of rat serum and colon tissue VIP,5-HT content decreased obviously.Ann intestinal antidiarrheal party maleic acid trimebutine high dose group and western medicine group differences between small,and Ann intestinal antidiarrheal low dose group can’t make the VIP,5-HT content decreased obviously.Conclusions:SD rats in senna lavage with bondage and under cold water stress can make higher sensitivity of internal organs,colic pain threshold is reduced,cause to change model of modern stimulation,coalition building methods cause intestinal disorders,and did not see the intestinal mucosa pathological changes,accord with the basic characteristics of IBS.Ann intestinal antidiarrheal party can reduce SD rats serum and intestinal mucosa of VIP,PYY,the content of 5-HT,and the ability to change the type of IBS-D rat visceral pain threshold,relieve the symptoms of IBS-D.Seen from the result,Ann intestinal antidiarrheal high dose group of the model group rats serum and colon VIP,reduce the effect of the content of 5-HT and maleic acid trimebutine difference is smaller,western medicine and Ann intestinal antidiarrheal low dose had little effect,so the dose Ann intestinal antidiarrheal big effect more apparent.
