Objectives:To explore the effect of LKB1 in epithelial-mesenchymal transition of hepatocellular carcinoma(HCC).Materials and Methods:We collected liver tissue samples from 90 patients with HCC at People's Hospital of Zhengzhou University. As the control of this study, liver tissue samples from 30 HBV-related cirrhosis cases and 30 hepatic hemangioma cases were analyzed. The expressions of LKB1,E-cadherin and N-cadherin were detected by using immunohistochemistry. Results:The positive LKB1 expression rate in HCC group was 31.1%(28/90),which was significantly lower compared with in HBV-related cirrhosis group(93.3%,28/30)and hepatic hemangioma group(96.6%,29/30).The positive expression rates of E-cadherin in HBV-related cirrhosis group and hepatic hemangioma group were 73.3%(22/30)and 66.6%(20/30)respectively,while only 45.6%(41/90)in HCC group.Compared with the low expression in HBV-related cirrhosis group(26.6%,8/30)and hepatic hemangioma group(13.3%,4/30),the N-cadherin expression rate was higher(70.0%,63/90)in HCC group. The expression level of LKB1 in HCC was associated with tumor's diameter, capsule, portal vein tumor thrombus and TNM-stage(p<0.05),but not with patients' age, gender, hepatitis B virus infection. The E-Cadherin expression was closely related with tumor's diameter, portal vein tumor thrombus and capsule(p<0.05).The N-cadherin expression was associated with capsule, portal vein invasion,TNM-stage(p <0.05).The positive expression rate of LKB1 in well differentiated or moderately differentiated HCC(36.99%,27/73)was higher than that in poorly differentiated or undifferentiated HCC(5.88%,1/17),and there was a statistically significant difference between them(P< 0.05).The expression of LKB1 was positively correlated with the expression of N-cadherin(p <0.05).Conclusion:The decreased degree of HCC differentiation is accompanied by the decreased degree of LKB1 expression. The LKB1 expression of HCC tissues is related with epithelial-mesenchymal transition.
