Objective To examine the expression of TLR9,My D88 and NF-?B m RNA in Peripheral blood mononuclear cells and the level of IL-6,TNF-? in the serum of patients with Henoch-Schonlein purpura.To compare the level of immune factor in the experimental group and control group and analyze the correlation of immune factor upstream and downstream.To reveal the possible role of signaling pathways of TLR9-My D88-NF-?B in immunological pathogenesis of Henoch-Schonlein purpura,Looking for upstream targets immune to provide basis for treatment of HSP.Methods Fourty HSP patients in acute phase were subsequently entered into study group who were collected in Department of Pediatrics,The second Affiliated Hospital of Zhengzhou University,form June 2015 to December 2016.Thirty-five cured HSP children in Outpatient department of Pediatrics constituted the control group.Real-time fluorescent quantitative polymerase chain reaction was used to measure the expression levels of TLR9,My D88 and NF-?B m RNA from Peripheral blood mononuclear cells.The levels of IL-6,TNF-? were measured in the serum of all subjects by ELISA.The Student t test was used for comparison between the HSP patients and the control.The one-way analysis of variance and LSD-t test were used to determine the differences among groups.The Pearson rank correlation analysis was used for the correlation among the expression levels of TLR9,My D88,NF-?B,IL-6and TNF-?.Results1.Compared with the control group,the expression levels of TLR9,My D88 and NF-?B m RNA from PBMC were up-regulated significantly in children with HSP(t = 5.68,P<0.01;t = 5.42,P<0.01;t = 112.95,P<0.01).2.Compared with the control group,the expression levels of IL-6,TNF-? in the serum were up-regulated significantly in children with HSP(t = 5.11,P<0.01;t=9.26,P<0.01).3.It is remarkably positive correlations with the expression levels of TLR9 and My D88,TLR9 and NF-?B,My D88 and NF-?B,NF-?B and IL-6,NF-?B and TNF-?.Conclusions1.The expression levels of TLR9,MyD88,NF-?B m RNA,IL-6 and TNF-? were remarkably increased in children with HSP.2.Our finding suggest that the expression levels of TLR9?MyD88 and NF-?B are positive correlations with the expression levels of immune downstream factor such as IL-6 and TNF-?.3.It is revealed that the signaling pathways of TLR9-MyD88-NF-?B has an important role in immunological pathogenesis of Henoch-Schonlein purpura.
