Objective:We set ovarian cancer models of nude mice with injection of SKOV3 cells firstly,in order to explore the antitumor effect of ginsenoside Rg1 on mice.Besides,we use ELISA to test serumal production of IL-12P40、IL-2、INF-? which can provide enhanced immunological basis for patients with ovarian cancer to cisplatin resistance and immune function of ginsenoside Rg1 combined with cisplatin in treatment.Methods:1.GroupingBALB/c nude mouse,female,are randomly averaged into 6 groups:Control group(no injection of SKOV3 cells)、low dose of ginsenoside Rg1、high dose of ginsenoside Rg1、cisplatin、combination of ginsenoside Rg1 and cisplatin、model group.The SKOV3 cells which have been cultured to acticity are inoculated under the left inguinal skin.After inoculation of tumor cells,the length and diameter of the tumor were measured every 2 days,then calculate tumor size.When the tumor volume reached 50mm3,the tumor size could be treated according to the corresponding treatment.The control group and the model group were given intraperitoneal injection of physiological saline,ginsenoside Rg1 small dose group,ginsenoside Rg1 high-dose group was given the corresponding dose of ginsenoside Rg1 gavage,cisplatin group was given intraperitoneal injection.2.Antitumor effect of ginsenoside Rg1After 14 days of medication,We use cervical dislocation to execute nude mice and removed tumor weighed to calculate tumor inhibition rate.According to the delivery days and tumor volume rendering the growth curve of tumor;tumor tissue will be made with HE staining.3.Investigation of mechanismWith ELISA detection of serum IL-12P40,IL-2,INF-? in nude mice serum collected after the last administration after 24 h.Results:1.Ginsenoside Rg1 low dose group,high dose group in day 4 and 6 compared with the model group,transplanted tumor volume less than the model group(p<0.05),in day 8,10,12,14,tumor volume decreased significantly compared with the model group(P<0.01);The cisplatin group,in day 4,6,8,10,12,and 14,the volume of transplanted tumor in nude mice was smaller than that in model group,and there was significant difference between the two groups(P<0.01);The combination group in day 4,6,8,10,and 12,the tumor volume was smaller than that in the model group(p<0.05),in the day 14,the tumor volume of combination group was significantly lower than that in the model group(P<0.01).2.Tumor growth curve:The slope of tumor growth curve of ginsenoside Rg1 was lower than that of model group,and the curve slope of high-dose group was lower than that of low-dose group;The tumor growth curve of cisplatin group was smaller than that of model group;The slope of curve of combination group was lower than that of model gruop,the growth curve of the combination group was smaller than that of cisplatin group.Chart 4.2.13.The inhibition rate: low-dose ginsenoside Rg1 group:29.07%,hige-dose ginsenoside Rg1 group:34.11%,cisplatin group:37.18%,combination group: 65.85%.The results were statistically significant(P<0.05).Table 4.2.24.There was no significant difference between the level of serum IL-2,INF-,IL-12P40 in drug treatment groups and that in model group(P>0.05);The serum IL-2,INF-?,IL-12P40 level in Cisplatin group was significantly lower than the model group(P<0.05).Table 4.3 and Chart 4.3.1、4.3.2、4.3.35.HE staining of subcutaneous tumor:In the model group,cells have different size and disorder arrangement,the cell structure is loose and chromatin is deeply stained which can be seen.There are many blood vessels in the stroma.In the ginsenoside Rg1 gruops,there was irregular necrosis in the parenchyma of the carcinoma,and the infiltration of immune cells was observed in the necrotic area.In the Cisplatin group,less lymphocytes and white blood cells infiltration were found in the necrotic areas.Conclusion:1.Ginsenoside Rg1 has antitumor effect in nude mice with ovarian cancer,but its effect is weaker than that of cisplatin.We can use it as a type of adjuvant antitumor drug.2.Ginsenoside Rg1 has synergistic effect on cisplatin.It can enhance the antitumor effect of cisplatinin in the treatment of ovarian cancer.3.Ginsenoside Rg1 may play a role in immune regulation in the treatment of ovarian cancer in nude mice: Rg1 promote anti-tumor immune cells secreting immune effect factors IL-2,INF-,IL-12P40,enhancing body anti-tumor immune response.4.Ginsenoside Rg1 may be used as an adjuvant with cisplatin in the treatment of ovarian cancer,reversing the body’s tolerance to chemotherapy so that chemotherapy completed on schedule.
