Objective:To observe the intervention of controlled hypotension in early reperfusion to PPAR-gamma mRNA caused by rat liver ischemia reperfusion and to explore its mechanism.Methods:Thirty clean level male SD rats were randomly divided into 3 groups(n = 10 each): sham operation group(group S),liver ischemia-reperfusion group(group I/R),controlling hypertension group(group CH).Sham operation group only free liver door,don't block the liver blood vessels;Liver ischemia-reperfusion blocking porta 60 min then 24 h reperfusion,ischemia-reperfusion injury model is set up;Controlling hypertension group(CH)blocking porta 60 min,in the early reperfusion femoral vein to give a mixture of monarkite and esmolol,controlling step-down to 70 MMHG,maintain a target blood pressure perfusion 10 min,drug withdrawal to blood pressure rise then 24 h reperfusion.Executed rats after 24 h,broken neck for brain tissue,the left side of the brain tissue paraffin section for pathological observation,on the right side of the brain by colorimetric method determination of superoxide dismutase(SOD)activity and malondialdehyde(MDA)content,rt-pcr detection of brain tissue ppar-gamma mRNA expression of relative.Results:Then group S brain structure no exception;CH group I/R group reduce degree of brain injury.Compared with S group,the MDA content of I/R group and CH group and ppar-gamma mRNA expression of relative significantly higher(P <0.05),SOD activity decreased significantly(P <0.05);CH group of MDA content in brain tissue and ppar-gamma mRNA expression of relative lower than I/R group(P <0.05),while SOD activity was significantly increased(P <0.05).Conclusion: Early reperfusion control pressure can reduce the rat liver ischemia reperfusion brain injury has certain brain protection,mechanism may be related to decreased MDA content and increase SOD activity,lower ppar-gamma mRNA expression and so on.
