Objective Establishment of experimental animal model of myocardial ischemia / reperfusion by ligation of the left anterior descending coronary artery in rats,and to investigate the effects of PPAR-γ on ischemia/reperfusion(I/R) rats with malignant arrhythmia in myocardial.Methods 24 SD rats were randomly divided into 4 groups,Each group consists of 6.There are: Sham group, I/R group, ROS group and GW group. Myocardial I/R injury was induced by ligation of the left anterior descending coronary artery, with ischemia for 30 min and reperfusion for 2 h. The whole process limb II lead electrocardiogram was applied to observe the frequency of malignant arrhythmia and record the corrected QT(QTc) interval changes; RT-PCR was used to detect the expression of PPAR-γ mRNA.Results There are 5 cases presented malignant arrhythmia in ROS group, 2 in I/R and 1 in GW, while Sham group is 0. ROS group has a prolongation of the QTc interval than the other groups after took out stitches(all of P < 0.05), the QTc interval is shorten in ischemia / reperfusion process in GW group than in I/R group and ROS group(all of P < 0.05). The expression of PPAR-γ mRNA significantly increased in other three groups than in Sham group(all of P < 0.05), ROS group has the highest expression level, and the expression of PPAR-γ mRNA reduced in GW group than in I/R and ROS(all of P < 0.05).Conclusion Over expression of myocardial PPAR-gamma after pretreatment by rosiglitazone,but over expression of PPAR-γ may lead to the occurrence of malignant arrhythmia in myocardial of ischemia/reperfusion rats.
