Objective: To disscus the hypoxia-inducible factor-1 alpha(HIF-1?)and vascular endothelial growth factor(VEGF)in the pathogenesis of HPH of neonatal rats by researching the mRNA and protein expression of HIF-1? and VEGF in HPH neonatal rats lung tissue and the relationship with the pulmonary artery pressure(PAP).Methods:According to the random number table method,the 80 wistar neonatal rats were divided into HPH group and control group,The rats of the two groups were put in the condition of hypoxia by 3,7,14 and 21 days and then 10 rats of HPH group and control group were picked up,Through testing the average pulmonary artery pressure(mPAP)to judge whether modeling success;Determining HIF-1? and VEGF mRNA expression level and protein expression level of HIF-1? and VEGF with Western Blot methods in lung tissue.Results: 1.On 3,7,14 and 21 day of hypoxia,the readings of mPAP on HPH groups which had a significant increase compared with control groups,the difference was statistically significant(P < 0.05).2.On 3,7and 14 day of hypoxia,the mRNA expression level of HIF-1a in the lung tissue on HPH group higher than the control group,the difference was statistically significant(P < 0.05);On 7,14 and 21 day of hypoxia,the mRNA expression level of VEGF in the lung tissue on HPH group higher than the control group,the difference was statistically significant(P < 0.05).3.On 7 day of hypoxia,the protein expression level of HIF-1? in lung tissue on HPH group there was statistically significant difference compared with control group(P <0.05);On 7,14 and 21 day of hypoxia,the protein expression level of VEGF in the lung tissue on HPH group higher than the control group,the difference was statistically significant(P < 0.05).Conclusion HIF-1? and VEGF in the occurrence and development of neonatal rat HPH. |
