Study On The Effective Substances And Quality Control Of Codonopsis Radix In Treating Pectoral Stuffiness And Pains

As we know,Dangshen is a tonic traditional Chinese medicine,China Pharmacopoeia 2015 version of the Dangshen is the dried root of Codonopsis pilosula(Franch.)Nannf.、Codonopsis pilosula Nannf.var.modesta(Nannf.)L.T.Shen or Codonopsis tangshen Oliv..Baitiao Codonopsis pilosula produced in gansu province is the main Codonopsis commodity.At present,the quality standard of Radix Codonopsis is not perfect,the traditional evaluation quality of Dangshen mainly by its sugar content to measure,"sweet plus",2015 version of the Chinese Pharmacopoeia has no dangshen content determination method,only Lobetyolin TLC qualitative identification and alcohol-soluble extract indicators.Some clinical and pharmacological studies that Codonopsis has a good therapeutic effect on angina pectoris of coronary heart disease,and the role is multi-faceted,multi-level.But there is no study on substance basis for the efficacy of Codonopsis treatment of chest pain disorder reported.Codonopsis pilosula contains a lot of fructose,glucose,sucrose,inulin and other sugar ingredients.In addition to sugar,the remaining extractIn(less than 5%)contained a large number of various types of polar compounds,are these molecules from Dangshen support the function of treating pectoral stuffiness and pains,so we hope to be able to find active compounds in Dangshen,through the control of these components to establish effective quality control method.In this paper,the research content mainly includes the following two aspects:(1)Study on the effective substances of Dangshen in treating pectoral stuffiness and pains.In this paper,the effects of different extracted parts of Codonopsis pilosula were screened by myocardial ischemia model in rats and dog hemodynamic experiments.Eight compounds were obtained from the effective fraction by using conventional extraction and separation techniques,such as macroporous resin,ion exchange resin,C18 reversed-phase filler and silica gel chromatography separation materials.By cell experiment,it was found that codonopyrrolidiums B had obvious protective effect on H2O2 damaged myocardial cells.Through the cell experiment and literature retrieval,which inferred the codonopyrrolidiums B,adenosine and vanillic acid may represent the effective substances of Codonopsis in the treatment of pectoral stuffiness and pains.(2)Study on quality control of active ingredients of Codonopsis pilosula.In this paper,high performance liquid chromatography(HPLC)was used to study the contents of codonopyrrolidiums B,adenosine and vanillic acid on the quality control of Codonopsis pilosula,and it was expected that the quality of Dangshen can be controlled more comprehensively,to ensure its clinical effect for the treatment of pectoral stuffiness and pains.The content of acetylcodonopyrrolidiums B in Baitiao Codonopsis pilosula was 0.299-0.443mg/g by high performance liquid chromatography.Codonopyrrolidiuns B is currently found only in the Codonopsis pilosula,has good specificity.The content of adenosine in Baitiao Codonopsis pilosula was 0.387-0.614mg/g by high performance liquid chromatography.It is reported that adenosine has the effects of coronary artery dilation and mediating ischemic preconditioning,reducing reperfusion injury and so on.The content of vanillic acid in Baitiao Codonopsis pilosula was 0.0136-0.0185mg/g by high performance liquid chromatography.Phenolic acids,such as vanillic acid,have long been considered as natural antioxidants,which can protect the cardiomyocytes from free radical damage.The content of total phenolic acid in Baitiao Codonopsis pilosula was 1.69-1.92mg/g by colorimetric method.Because of the variety of phenolic acids in Codonopsis pilosula and the low content of single component,the content of total phenolic acid was determined by potassium ferricyanide and ferric chloride colorimetric method.
