Objective: In order to discuss the regulation mechanism of athletes ’energy metabolism in muscle and amino acid intervention,we should establish a model of exercise to explain the mechanism of the structure and function of athletes’ myocardium.Beclin-1 and LC-3II as a key protein to measure autophagy activity can be negatively regulated by m TOR,whereas AMPK promotes autophagy by inhibiting m TOR,and AMPK and m TOR synergistically regulate the energy metabolism of the myocardium.Methods: Eighty male SD rats were randomly divided into four groups: the experimental group,the rats were divided into four groups according to the principle of randomized control.Control group(Con)、Glucose alone(G),Leucine group(L),Glucose and Leucine group(G+L),Exercise Group(E),Exercise Glucose(E+G),Exercise Leucine(E+L),Exercise-supplemented glucose-leucine group(E+G+L).The expression of LC3-II,Beclin-1,P-MTOR,MTOR,P-AMPK and AMPK in myocardium was detected by western blotting.The experimental data were analyzed by immunohistochemistry.Using SPSS22.0 two-way analysis of variance for statistical processing.Results:(1)The expression of autophagy protein LC-3II and Beclin-1 in cardiomyocytes was consistent,and the control group was lower than that of exercise group,glucose supplement group and exercise supplement group(P <0.05);And the amino acid supplementation did not improve the autophagic activity.The results showed that moderate-intensity exercise and glucose supplementation could promote the expression of autophagic protein and amino acid supplementation did not improve autophagic activity;(2)Supplementation of glucose and leucine can inhibit the activity of m TOR,activity 24 h after exercise intervention m TOR activity returned to normal;(3)The activity of AMPK was not active in 24 hours after exercise and glucose and leucine groups.Conclusions:(1)moderate-intensity exercise training can increase the expression of autophagy protein Beclin-1 and LC3-II;(2)There was no significant difference in the expression of autophagy of myocardium in rats with the same amount of glucose and leucine.
