Astragalus Plus Zedoary With The Combination Of Oxalipl-Atin Influence The Expressions Of Chemokines And Chemo Kine Recepters In CT26.WT Tumor Orthotopically Mice

[Objective]Establish BALB/c mouse model of CT26.WT orthotopic implantation to investigate the effect of Astragalus Plus Zedoary(APZ)extract combine oxaliplatin(L-OHP)formula on murine colorectal cancer cell line CT26.WT inhibition of tumor metastasis,regulate protein and mRNA expressions of CXCR3,CCR6,CXCL9,CCL20.[Methods]1.BALB/c mouse model of CT26.WT orthotopic implantation was established at the first time.Observe CRC tumor growth and liver metastasis of the mouse model.2.35 mouse was divided into 5 groups:control group,L-OHP group(5mg/kg),combination of L-OHP(5mg/kg)and APZ(with three different doses:6,3,1.5g/kg).After 21 days of treatment,investigate weight,orthotopic tumor weight,liver metastasis of each mouse.3.By Real-time PCR,detect the expression of CXCR3,CCR6,CXCL9,CCL20 gene in tumor and liver tissue after three weeks treatment.4.By Western Blotting,detect the expression of CXCR3,CCR6,CXCL9,CCL20 protein in tumor and liver tissue after three weeks treatment.[Results]1.Successfully establish BALB/c mouse model of CT26.WT orthotopic implantation at the rate of 100%.2.The use of L-OHP and combination groups can inhibit tumor growth,L-OHP group was better than all three L-OHP+APZ groups(P<0.01).L-OHP+APZ groups can decrease the chance of liver metastasis(P<0.05).3.The mRNA expressions of CXCR3 and CCR6 decreased in the model group and low-dose group compared with the control group(P<0.01),especially the effect of L-OHP combind with low dose of APZ were stronger than other combination therapies(P<0.01).CXCL9 and CCL20 gene expressions haven’t show statistical significance in four medication groups.4.Compared with the control group,CXCR3 and CCR6 protein expressions were decreased remarkably in all four medication groups(P<0.05 or P<0.01).With the use of L-OHP+APZ,CXCL9 and CCL20 protein were inhibit compared with the control group in liver tissues(P<0.01).[Conclusion]L-OHP+APZ treatment reduce the liver metastasis in CT26.WT orthotopic implantation mouse model significantly,inhibit the protein and mRNA expressions of CXCR3 and CCR6 depending on the dose of APZ,when concentration was 1.5g/kg,CXCR3 and CCR6 protein inhibited significantly.The trend of CXCR3 and CCR6 mRNA expressions were same as their protein expressions.L-OHP+APZ reduce CT26.WT liver metastasis might associated with decreased protein and mRNA of CXCR3 and CCR6.After L-OHP+APZ treatment,CXCL9 and CCL20 gene and protein expressions still need more experiment.
