PURPOSE To investigate the expression and clinical significance of in glioma and explore the potential of the 4 CTAs serving as immunotherapeutic target for glioma.METHODS Expression of the ACTL8, CTCFL, OIP5 and XAGE3 four CTAs protein was detected by immunohistochemical staining in 108 pathologically diagnosed glioma cases. Statisical analyses of the correlation between four CTAs expression and Clinical pathological features of glioma cases was conducted by SPSS 17.0 statistics analysis software.RESULTS(1) The expression rates of ACTL8 protein in glioma were 21.3%(23/108 ) , the expression rates of ACTL8 protein in low grade glioma (WHOⅠ-Ⅱ grade) were 25.0%(11/44),and the expression rates of ACTL8 protein in low grade glioma(WHO Ⅲ-Ⅳgrade) were 18.8%( 12/64).Aomg the ACTL8 protein positive glioma specimens, 14 of them show low staining, 6 show medium staining and 3 high staining. The expression of ACTL8 protein in glioma have correlation with the sex,KPS(P<0.05).(2) The expression rates of CTCFL protein in glioma were 34.3%(37/108), the expression rates of CTCFL protein in low grade glioma(WHO Ⅰ - Ⅱ grade) were 29.5%( 13/44), and the expression rates of CTCFL protein in low grade glioma(WHO Ⅲ - Ⅳ grade)were37.5%(24/64). Aomg CTCFL protein positive glioma specimens,17 of them show low staining, 13 show medium staining and 7 high staining. No correlation was found between the expression of CTCFL protein in glioma and clinical pathological parameters (P<0.05).(3) The expression frequencies of OIP5 protein in glioma were 19.4%(21/108), the expression frequencies of OIP5 protein in low grade glioma(WHO Ⅰ - Ⅱ grade) were 29.5%( 13/44), and the expression rates of OIP5 protein in low grade glioma(WHO Ⅲ-Ⅳ grade) were 2.5%(8/64).Aomg OIP5 protein positive glioma specimens, 14 of them show low staining, 5 show medium staining and 2 high staining. The expression of ACTL8 in glioma have correlation with the WHO grade, KPS(P<0.05),no correlation was found between the expression of OIP5 protein in glioma and others clinical pathological parameters(P< 0.05).(4) The expression frequencies of XAGE3 protein in glioma were 23.2%(25/108), the expression frequencies of XAGE3 protein in low grade glioma(WHO Ⅰ -Ⅱ grade) were 22.7%( 10/44), and the expression frequencies of XAGE3 protein in low grade glioma(WHO Ⅲ-Ⅳ grade)were 25.9%( 15/64). Aomg XAGE3 protein positive glioma specimens,14 of them show low staining, 6 show medium staining and 3 high staining. The expression of XAGE3 protein in glioma have correlation with KPS(P<0.05), no correlation was found between the expression of OIP5 protein in glioma and others clinical pathological parameters(P<0.05).(5) 41.7% (45/108) of the specimens do not express any kind of CTAs.28.7% (31/108) of the specimens express one of the CTAs. 19.4%(21/108) of the specimens express two CTAs. 10.2% (11/108) expressed the three kinds of CTA synchronously.CONCLUSION ACTL8, CTCFL, OIP5 and XAGE3expressed in glioma, in which CTCFL have highest expression frequency, followed by XAGE3, OIP5 and ACTL8. Co-expression of these four CTAs provides a new insight for improving the overall effect of tumor immunotherapy.The patients with CTCFL protein positive have poor prognosis compare to those with CTCFL protein negative, suggest than CTCFL may be save as a poteintially prognostic biomarker.
