Objective:Human papillomavirus(HPV)infection is not the only factor that leads to the development of cervical intraepithelial neoplasia(CIN)and cervical cancer(CC).They are the common result of multi-factors such as behavior risk factors,genetic susceptibility factors,environment factors,and condition of body immunity and nutritional.In this study,we examined the levels of folic acid and Thl/Th2 cytokines in peripheral blood of patients with cervical cancer and cervical intraepithelial neoplasia.Through the experiment,we made an analysis on factors which influenced occurrence and progression of cervical lesions from the level of trace elements and the level of immunity.Thus,the study provided a new idea for the prevention,detection and treatment of cervical precancerous lesions and cervical cancer.Method:All the samples were collected from women attending the Liuzhou Worker's Hospital,and Liuzhou Maternal and Child Health-Care Hospital from October 2015 to October 2016.We selected patients of chronic cervicitis(CI,28),cervical intraepithelial neoplasia ?(CIN ?,54),cervical intraepithelial neoplasia ?(CIN ?,24),cervical intraepithelial neoplasia III(CIN III,36),and cervical cancer(CC,35)as the research object of our study,they were experienced thinprep cytologic test(TCT),colposcopy examination screening,high risk HPV detection,and finally diagnosed with pathology.We determined serum folic acid of research objects by using the method of direct chemiluminescentce immunoassay,and erythrocyte folate by using the method of electrochemiluminescence.Flow cytometry(CBA)was used to detect the related cytokines(L-2,IL-4,IL-6,IL-10,IFN-y).Finally,we conducted regression analysis by using SPSS 17.0 software for chi-squared test,trend chi-square test,Kruskal-Wallis H test and t-test.Results:1.The infection rate of high risk HPV in CIN I group,CIN II group,CIN III group,CC group was higher than that in CI group,the difference was statistically significant(P<0.05),and with the aggravation of cervical intraepithelial neoplasia,the risk of HPV infection gradually increased(trend x2=25.8,P<0.05).2.The serum folic acid content of CI group(37.42±4.55 nmol/1)was higher than that of CIN I group(23.68±4.51nmol/l),CIN II group(17.92 ± 3.91 nmol/1),CIN III group(15.95 ± 5.0lnmol/l),and CC group(12.91±2.31 nmol/1);erythrocyte folate levels in the CI group(2313.51 ± 65.6 nmol/1)were higher than that of CIN group ?(1892.86 ± 47.2 nmol/1),CIN??group(1652.35 ±65.7 nmol/1),CIN ? group(1583.47 ± 52.7 nmol/1),and CC group(1481.51 ± 36.5 nmol/1).Significant differences were shown in the distribution of serum folic acid and erythrocyte folate in each group(H=128.61,P<0.05;H=10.07,P<0.05).The result analysis of trend chi-square test showed that the OR value of serum folic acid analysis in CIN I group and CIN II group had no significant difference(P>0.05).With other groups' decrease in the level of serum folate,the OR in CIN III group and CC group showed an upward trend,and the OR value all greater than 1(P<0.05);in the analysis of erythrocyte folate level,the OR value of each experimental group were statistically significant(P<0.05).With the decrease of erythrocyte folate level,the OR value in CIN ?,CIN ?,CIN ? group and CC group showed an upward trend.3.The serum levels of IL-2,IL-4,IL-6,IL-10 and IFN-y in each experimental group were compared with CI group's.The levels of IL-2,IL-4 and IFN-? were not statistically significant(P>0.05);the serum level of IL-6 is higher than experiment group(P<0.05);the serum level of IL-10 only in CIN ? group and CC group were significantly different from those in control group(P<0.05),which were higher than those in the control group.Conclusions:1.High risk HPV infection is the risk factor for the occurrence of CIN and cervical cancer.2.Low folate levels were associated with CIN and cervical cancer.Folate deficiency maybe increases the risk of CIN and cervical cancer 3.The levels of serum IL-6 and IL-10 were significantly increased in patients with cervical intraepithelial neoplasia and cervical cancer,suggesting that the body may be in a state of immunosuppression.The determination of IL-6 and IL-10 has great significance in the diagnosis and treatment of CIN and cervical cancer.
