The Influence Of Tong Xie Yao Fang On 5-HT,CGRP And VIP In Serum Of Diarrhea-predominant Irritable Bowel Syndrome Model Rats

Objective: Diarrhea predominant irritable bowel syndrome(IBS)is one of the most common functional gastrointestinal disorder disease,traditional Chinese medicine has achieved good clinical efficacy in treating this disease through the dialectical treatment,and by using the Tong Xie Yao Fang(TXYF).However,at present,it is still necessary to explore and study the principles and mechanisms of traditional Chinese medicine-TXYF in treating the disease.In order to get the therapeutic effect and possible mechanism of TXYF in treating IBS,in this academic degree dissertation,experiment results has been get in the process of observing the effect of TXYF on the index of gastrointestinal hormones,IBS-D rats,5-HT,CRGP and VIP.Methods: SD rats were randomly divided into control group and IBS-D model group.In the model group,the model was made by combining the method of chronic unpredicted mild stress and restriction.After the modeling,the model group were randomly divided into control group and pinaverium bromide treatment group,and TXYF low dose,meta-dose,high dose treartment group.The control group was fed normally without special treatment.After the treatment,all the SD rats were collected blood by the heart punctures,then the serum were tested with 5-HT,CGRP and VIP were detected by ELISA.The measurement of carbon propulsive distance and calculated carbon advance rate were made after all the processes.Results: 1.General situation of SD rats: before modeling: the control group mental health is fine,their reaction to light stimulation were quick-minded,food intake and drinking had no obvious change,their had smooth white fur and their faecal pellet is large and moist.After modeling: Rats in model group had spiritual burnout,however their were particularly sensitive to various external stimuli and irritability,food intake was reduced than before and had less drinking,rats’ fur turned into owe smooth and dark yellow,faecal pellet presented watery stool,or large granular and with high liquid content there were some unformed shit stick at anus.After treatment: the rats were in good spirit,and their response to light stimulation were quick-witted,the food intake and drinking had improved,silky white tends to be full.Fecal pellet formation or thinning were reduced.2.Body weight change in SD rats: after modeling,Compared with the control group,there were less weight gain in model group(P=0.00,P<0.01).3.After drug(TXYF)treatment: The SD rats in the treatment group increased more than the model group(P<0.05).3.Results of IBS-D model SD rats:(1)Weight change: the SD rats in model group were lower than control group(P<0.05).(2)evaluation of stool characters: the score of the model group was higher than control group(P<0.01).4.Analysis of the changes of serum in 5-HT,CGRP and VIP:(1)statistical analysis of 5-HT concentration in SD rats: TXYF meta-dose and high dose group will increase the content of 5-HT in serum(P<0.01),TXYF low dose group and pinaverium bromide group had no significant change(P>0.05).Compared with other treatment groups,the best therapeutic effect was in TXYF high dose group(P<0.01),the low,meta and high dose of TXYF had dose effect relationship with 5-HT in serum of SD rats.(2)SD rat CGRP concentration statistical analysis results: TXYF low,medium and high dose group will increase the content of CGRP in serum(P<0.01),there were no significant difference between the low dose TXYF group and pinaverium bromide treatment group(P>0.05).Compared with the other treatment groups,the best treatment effect was in TXYF high dose group(P<0.01),the low,middle and high dose of TXYF had a dose effect relationship with the concentration of CGRP in serum of SD rats.(3)statistical analysis of VIP concentration in SD rats: TXYF low,high dose group could reduce the content of VIP in serum(P<0.01),TXYF middle dose group and pinaverium bromide treatment group had equivalent therapeutic effect(P>0.05).Compared with other treatment groups,the best therapeutic effect was in the high dose group(P<0.01),The low,meta and high dose of TXYF had dose effect relationship with VIP in serum of SD rats.Conclusion: 1.The modeling method of the combination of chronic unpredicted mild stress and restriction is economical and effective,and proved to be capable in simulation of liver depression symptoms of spleen deficiency and cardinal symptom of IBS-D which regard as the SD rats model of liver depression of traditional Chinese medicine.2.There were significantly improvement of 5-HT,CGRP and VIP in serum of SD rats in modeling group.3.The possible approach of treatment in IBS-D of TXYF is through regulating and controlling the concentration of 5-HT,CGRP and VIP in serum of SD rats.TXYF proved to relieve IBS-D cardinal symptoms in this experiment and is better than pinaverium bromide which illustrate TXYF is effective in treating IBS-D.
