Objective: TCAB1(telomerase cajal body protein 1,TCAB1 also denoted WRAP53 or WDR79)has been noticed as a natural antisense transcript that regulates the P53 tumor suppressor,which can stable P53 mRNA,maintenance telomere and impair DNA damage.Many research previously identified TCAB1 is overexpressed in a variety of cancer cell lines of different origin and that TCAB1 overexpression was significantly correlated with tumor invasion,clinical stage,lymph node metastasis and prognosis transformation.P53 is a kind of apoptosis regulation gene,which is related to inducing apoptosis of tumor cells.However,it has been reported that Knockdown of the TCAB1 protein triggers massive apoptosis.The adjacent esophageal squamous cell carcinoma tissues were compared and we detected the expression of TCAB1 and P53 protein in 160 esophageal squamous cell carcinoma tissues,the expression of TCAB1 and P53 mRNA in52 esophageal squamous cell carcinoma tissues and analyzed the correlation between TCAB1 and P53.At the same time,we analyzed the above results with age,stage,degree of differentiation,lymph node metastasis,tumor size and so on in order to investigate the occurrence of TCAB1 in esophageal squamous cell carcinoma and its role in the development.Methods: Immunohistochemistry(IHC)was used to evaluate expression of TCAB1 and P53 protein in160 ESCC tissues and the corresponding nonneoplastic esophageal epithelium tissues(including 52 pairs of freshtissues),and correlate TCAB1 and P53 protein expression with clinicopathological parameters of ESCC patients,and explore the correlation between the two proteins.Real time quantitative polymerase chain reaction(RT-qPCR)was used to detect the expression of TCAB1 and P53 mRNA in 52 ESCC tissues and the corresponding nonneoplastic esophageal epithelium tissues,and correlate TCAB1 and P53 gene expression with clinicopathological parameters of ESCC patients,and explore the correlation between the two genes.Results: TCAB1 has been detected in ESCC and nonneoplastic esophageal epithelium,and Both TCAB1 protein and mRNA levels were upregulated in ESCC tissue compared with adjacent nonneoplastic esophageal epithelium(P < 0.05).By IHC,The expression of TCAB1 protein was correlated with tumor invasion depth(X2=5.77,P=0.016),but not age,gender,clinical stage,tumor differentiation,lymph node metastasis and tumor size.P53 has been detected in ESCC and nonneoplastic esophageal epithelium,and Both P53 protein and mRNA levels were upregulated in ESCC tissue compared with adjacent nonneoplastic esophageal epithelium(P<0.05).By IHC,The expression of P53 protein was correlated with tumor size(X2=4.26,P=0.039),but not age,gender,clinical stage,tumor differentiation,tumor invasion depth and lymph node metastasis.Spearman method was used to analyze the correlation between two proteins,and the difference was not statistically significant(r=0.088,P=0.367).Spearman method was used to analyze the correlation between two genes,and the difference was not statistically significant(r=-0.135,P=0.581).Conclusion: The expression of TCAB1 protein level in esophageal squamous cell carcinoma was higher than that in adjacent normal mucosa,which correlated with tumor invasiveness and can be used as an evaluation elements for the transformation and prognosis of esophageal squamous cell carcinoma.The expression of P53 protein level in esophageal squamous cell carcinoma was higher than that in adjacent normal mucosa,which was related to tumor size and can be used as an evaluation elements for tumor progression.The expression of TCAB1 and P53 mRNA in esophageal squamous cell carcinoma was significantly higher than that in normal mucosa adjacent to carcinoma,suggesting that the expression of TCAB1 and P53 mRNA may be related to the occurrence of esophageal squamous cell carcinoma.In esophageal squamous cell carcinoma,bothTCAB1 and P53 in protein and mRNA levels has no correlation.
