ObjectiveBased on the discovery of the total flavonoids extract of Albizia julibrissin with sedative and hypnotic effects,through the understanding of the complexity of traditional Chinese medicine system,combined with the previous experimental results,In this study,high-performance liquid chromatography(HPLC)was used to establish the fingerprints of total flavonoids from Albizia julibrissin in different areas.The gray correlation theory was used to combine the fingerprints of traditional Chinese medicine with pharmacodynamics.To elucidate the correlation between the fingerprints of the total flavonoids extract of Albizia julibrissin and the sedative and hypnotic efficacy,and to characterize the pharmacological basis of the medicinal materials of Albizia julibrissin.MethodsStudy on HPLCfingerprint of total flavonoids extract from Albizia julibrissin:According to the research group set up in the early extraction process of total flavones from Albizia julibrissin,preparation of extracts of total flavones from Albizia julibrissin test using high performance liquid chromatography(HPLC)on 12 lots of different origin analysis of total flavone extract of Albizia julibrissin,The determination of wavelength,mobile phase composition and gradient elution conditions were carried out by using quercitrin standard as reference substance,and the methodological investigation was completed.The establishment of 12 batches of different origin of the total flavonoids extract fingerprints according to the requirements4of chemical fingerprints,the application of the National Pharmacopoeia Commission promulgated the "Chinese medicine chromatographic fingerprint similarity evaluation system software(2004A version)" 12 batches of different origin of Albizia julibrissin The fingerprints of total flavonoids were evaluated by similarity.Albizia julibrissin flavonoids extracts the curative effect evaluation of insomnia rats:In this study,intraperitoneal injection of PCPA method was used to make the model of insomnia rats,and the effect of the total flavonoids extract of Albizia julibrissin on the treatment of insomnia rats was confirmed by the pentobarbital sodium sleep co-experiment.Albizia julibrissinflavonoids extracts sedative-hypnotic mechanism research: Making insomnia rat model,HE staining and immunohistochemical staining.The positive expression of GABA in the hypothalamus and cerebral cortex of rats was taken as an index,to study the sedative and hypnotic mechanism of total flavonoids extract from Albizia julibrissin in different batches.Study on the spectrum-activity relationship in sedative and hypnotic effects of Albizia julibrissin flavonoids extract: The relative peak area values of the characteristic peaks of the fingerprints of 12 batches of different origin Albizia julibrissin flavonoids extractwere normalized and the ratio of Glu /GABA reduction in pharmacodynamic experiments,using the gray relational analysis method,calculate the size of the chemical constituents represented by the common characteristic peaks of the fingerprints on the contribution of sedative hypnosis.ResultsIn this study,the fingerprints of the total flavonoids extracted from 12 batches of different habitats were established.The results showed that the total flavonoids of Albizia julibrissin in different habitats had enhanced theinhibitory neurotransmitter GABA positive in hypothalamus and cerebral cortex of insomnia rats The role of expression.Based on Albizia julibrissin flavonoids extract of different regions fingerprint in 19 of characteristic peak and the correlation of sedative hypnotic effect research,get the fingerprint the chemical composition of the common characteristics of the peak represents the contribution of sedative hypnotic effect sequence is: X9>X15>X12>X18>X8>X17>X10>X5>X1>X7>X13>X3>X11>X14>X2>X19>X6>X4>X16.ConclusionsIn this study,the fingerprints of total flavonoids extracted from 12 batches of different habitats were established,and the insomnia model was established by intraperitoneal injection of PCPA method.The mechanism of sedative and hypnotic effects of total flavonoids of Albizia julibrissin was studied.The results showed that the total flavonoids extract of Albizia julibrissin had the effect of sedation and hypnosis through the synergistic effect of its internal "effective component group".This study only identified a part of the sedative and hypnotic effect of the larger contribution to the structure of the composition.As the composition of traditional Chinese medicine is very diverse,the confirmation of the effective ingredients of the material basis needs to be further studied and perfected.Through the study of this subject,in order to expand the clinical application of Albizia julibrissin,to clarify the basis of its pharmacological material provides a theoretical and experimental basis.
