| Objective:To evaluate the quality of different enterprises Yaotong tablets by establishing two kinds of methods of content determination and fingerprint. We establish a kidney yang deficiency model with male mice, to carry out a study about healing kidney yang deficiency model with Yaotong tablets. The results of pharmacodynamics test rational of fingerprints to study the relationship between Yaotong tablets HPLC fingerprint and efficacy of treating kidney yang deficiency mice.Methods:First of all, a method was developed to assay Yaotong tablets by HPLC and GC chromatography, it was used to analyze the content of the sample. We determine the content of four major components (paeoniflorin, asperosaponin VI, psoralen and isopsoralen) in Yaotong tablets by high performance liqulid chromatography. We determine the content of two major components (ligustilide, cinnamic aldehyde) in Yaotong tablets by gas chromatography. The data of experiment will be treated by software SPSS, to explore sample problems of different enterprises Yaotong tablets in markets. Ones again, the similarity of sample fingerprints were used to analysis sample quality by HPLC and gas chromatography. Basing on the study of fingerprints the Yaotong tablets pharmacodymics. Yaotong tablets of the different enterprises were treated male mice with kidney yang deficiency induced by hydrocortisone to study pharmacodymics indexes, these indexes include automomic activity, analgesic latency time, drowning time, weight gain, testing the fingerprints whether it was reasonable as a standard to evaluate the quality of the sample. Finally, partial least squares were applied to establish relationship between fingerprint and pharmacodynamics. We find out the correlation between the test indexes and the chromatographic peak. It further clarifies the material basis for efficacy of Yaotong tablets.Results:Two kind of chromatography measured content of the active components in different enterprises Yaotong tablets, these included paeoniflorin, asperosaponin VI, psoralen and isopsoralen, ligustilide and cinnamic aldehyde. We find the significant differences of content in between batches in different enterprises. HPLC fingerprint and GC fingerprint evaluate the whole quality of Yaotong tablets. It finds that they are basically the same to evaluate quality of Yaotong tablets between pharmacodymics indexes and evaluation of fingerprint system, and find out a relationship between fingerprint and pharmacodymics indexes.Conclusion:There are many differences on the overall quality about Yaotong tablets, the methods for determination of the content and for fingerprint are credible, it can provide the improvement reference of quality standard of Yaotong tablets. |
