Objective To investigate the effects of dequalinium chloride on apoptosis and proliferation and invasion of glioma cells.Methods With 50?mol/L,100?mol/L,200?mol/L of dequalinium chloride role in glial tumor cells U251,U87,C6,action time respectively for 12 h,24h,48 h,72h,96 h.Cell proliferation was detected by MTT assay.Flow cytometry was used to detect cell apoptosis after 76?mol/L dequalinium chloride 48 h.Transwell was used to detect cell invasion after 76?mol/L dequalinium chloride 48 h.Western blot detection of cells in PI3K?p-PI3K?Akt?p-Akt expression levels.Results 50?mol/L,100?mol/L,200?mol/L of dequalinium chloride has inhibitory effect on U251,U87,C6 glioma cell proliferation.Dequalinium chloride on glioma cell proliferation inhibition with increasing reaction time increases,increases with the increase of drug concentration.IC50 of dequalinium chloride role in glioma U251,U87,C6 cells 48 h in descending order was: C6>U87>U251.The apoptosis rate of glioma cells after 76?mol/L dequalinium chloride was significantly higher than the control group(P<0.01).The invasion ability of glioma cells after 76?mol/L dequalinium chloride was significantly lower than the control group(P<0.01).The expression of PI3 K and Akt in dequalinium chloride group and control group showed no significant difference,and p-PI3 K,p-Akt expression level decreased significantly.Conclusions Dequalinium chloride on glioma cell proliferation and invasion has inhibitory effect,has a promoting effect on glioma cell apoptosis,the mechanism of action related to PI3 K / Akt signaling pathway.
