ObjectiveTo study the functional recovery of recombinant human erythropoietin and?-aescin after spinal cord injury in rats and the neuronal apoptosis in injured group.MethodsA total of 50 female SD rats were divided into five groups according to their injury and treatment: blank group,injury group,rh EPO group and ?-SE group,combined treatment group and blank group were treated with equal volume of saline intraperitoneal injection,In the injured group,the recombinant human erythropoietin was taken from the rh EPO group.The ?-SE group was treated with ?-aescin,and the combined treatment group was treated with recombinant human erythropoietin and ?-aescin,The BBB scores of each group were evaluated.NF-?B immunohistochemical results,tissue Ca2+ and Mg2+ contents were compared.Histological HE staining results were compared and immunohistochemical results were compared.Results?1?The BBB motor function scores of the combined treatment group were significantly higher than those of rh EPO group,?-SE group and injury group?P <0.01?.Rh EPO group and ?-SE group were slower than those in the injury group,but all of them were significantly lower than those in the treatmentgroup?P <0.05?.?2?blank group of animals with normal neurological function,normal exercise.The neurological function of the combined treatment group was better than that of rh EPO group and ?-SE group,the difference was statistically significant?P <0.05?.rh EPO group,?-SE group,combined treatment group had better neurological function than injury group.?3?The expression of NF-?B was also observed in the combined treatment group,and the trend was similar to that of the injury group,which was significantly different from the blank control group?P <0.05?.Rh EPO group,?-SE group was significantly lower than the injury group,the difference was significant,Which was significant at 8h and 14h?P <0.01?.?4?The content of Ca2+ in the blank control group after spinal cord injury was significantly higher than that in the injury group,rh EPO group,?-SE group and the combination group?P<0.05?.The Ca2+ content in the combined treatment group was significantly higher In the rh EPO group,?-SE group,the difference was significant?P<0.05?.The content of Mg2+ in the blank control group was significantly lower than that in the rh EPO group,the rh EPO group,the ?-SE group and the combination group?P <0.05?.The Mg2+ content in the combined treatment group was significantly lower than that in the rh EPO group,The difference was significant?P <0.05?.?5?rh EPO group and ?-SE group showed similar,but the degree of reduction,the combination of treatment group to improve more obvious.The area of lesion area was smaller than that of rh EPO group and?-SE group.There were hemorrhagic necrosis,hemorrhagic edema,less inflammatory reaction,less glial hyperplasia,small voids and less formation In the treatment of A,B two groups,the number of residual neurons more normal morphology,nucleolus clear,visible Nissl body,white matter within the nerve fibers arranged in a dense order.?6?The expression of bcl-2 in rh EPO group,?-SE group and combination therapy group was significantly higher than that in rh EPO group and ?-SE group?P<0.05?,and there was significant difference between the two groups?P<0.05?.In addition,the expression of fas positive apoptotic cells was higher in the injured group,and it was widely distributed inthe gray and white matter of the spinal cord.The expression of fas positive apoptotic cells was lower in rh EPO group,group B and combination therapy group,the difference was significant?P<0.05?.The levels of bcl-2 in the combination group were significantly higher than those in the rh EPO group,?-SE group and the combined treatment group?P <0.05?.?7?The distribution of dark blue to blue-purple apoptotic cells was observed in the spinal cord tissue sections of injury group,?-SE group and rh EPO group.A large number of apoptotic cells,?-SE group and rh EPO group were observed in the injury group.There was no distribution of apoptotic cells in the combined group.?8?The rate of apoptotic cells in the injured group,?-SE group and rh EPO group was significantly higher than that in the combined group,the injury group and the ?-SE group,the rh EPO group and the combined group?P <0.05?There was no significant difference between the ?-SE group and the rh EPO group?P> 0.05?.ConclusionsThe combination of recombinant human erythropoietin and ?-aescin is helpful to improve the BBB motor function score and promote the recovery of motor function in rats.?2?can prevent the local Ca2+ agglomeration caused by overload caused by SCI on nerve cells secondary injury,prevent the concentration of Mg2+ decreased,effectively improve the local Ca2+ overload and energy metabolism,delay the development of secondary nerve cell injury.?3?can improve the pathological changes of spinal cord injury in rats,reduce spinal cord neuronal apoptosis,can reduce secondary spinal cord injury.
