Objective: To investigate the effects of ERCC1-sh RNA targeting silencing excision repair cross-complementing genome 1(ERCC1)on the proliferation and apoptosis of lung cancer cell line A549/DDP cells treated by different concentrations of cisplatin.Methods: Human interfering RNA(RNAi)targeting human ERCC1 gene was constructed and used to transfect A549/DDP cells by lipofectamine 2000.The expression of ERCC1 in A549/DDP was evaluated by Real-time PCR and Western Blotting.The cell proliferation inhibition rate was assessed by MTT.Cell cycle and apoptosis were assessed by flow cytometry.Results: ERCC1-shRNA was successfully constructed and transfected into A549/DDP cells,both ERCC1 m RNA and protein expression were significantly down-regulated in A549/DDP cells transfected with ERCC1-sh RNA(P<0.05).The proliferation inhibition rate of ERCC1-sh RNA transfection cells was significantly increased compared to the negative control group and the blank control group.Furthermore,the IC50 value was decreased markedly,the cell cycle was arrested in G0/G1 phase and the apoptosis rate was increased(P<0.05).Conclusion: ERCC1-sh RNA inhibits the proliferation of A549/DDP cells and enhances cell apoptosis.
