| Objective:Detecting the typical compounds in exhaled breath of patients with liver failure and healthy volunteers by Enclosed EESI-MS,exploring the relationship and difference with metabolism and the serum biochemical indexes of liver failure.Method:1、Gathered the exhaled breath of patients with liver failure(n = 23)and healthy controls(n = 25),detected by Enclosed EESI-MS.2、Analyzed the original data by the partial least squares method(PLS),and selected the variables with significant difference.And then Searching for the high resolution molecular mass which corresponding to differential variables in the first order spectrum.3 、 Identified the high resolution molecular mass by two stage tandem mass spectrometry.Determined what compounds the high resolution molecular maybe and found their association with liver failure.4、SPSS17.0 software was used to analyze the molecular mass of the compounds in exhaled breath with the biochemical indexes of liver failure such as alanine aminotransferase(ALT),aspartate aminotransferase(AST),total bilirubin(TB),direct bilirubin(DB),prothrombin time(PT),prothrombin time activity(PTA).Result:1、It was revealed that exhaled breath of healthy volunteers and patients with liver failure were well differentiated by PLS,and 24 variables with the greatest difference were selected by using the model variables VIP value.2、Two stage tandem mass spectrometry was used to identify the high resolution molecular mass,and ascertain 10 compounds:isobutene(m/z 57),acetone(m/z 59),acetaldehyde(m/z 61),isoprene(m/z 61),2-butanone(m/z 73),cyclopentanone(m/z85),2,5-dimethylfuran(m/z 97),2-ethylacrylic acid(m/z 101),6-methylhexan-2-one(m/z 115),octanal(m/z 129).3、It was found that the serum biochemical index of liver failure was linearlycorrelated with the molecular mass of compounds in exhaled breath.Conclusion:1、There are dismilar compounds between the exhaled gas of healthy volunteers and patients with liver failure,which identified 10 species.For isobutene,acetone,acetaldehyde,isoprene,2-butanoneand and cyclopentanone,we can infer the metabolic process in vivo and the connection with disease.More studies are needed for 2-ethylacrylic acid,octylaldehyde and 6-methylhexan-2-one.2、The molecular mass was positively correlated with the severity of liver failure.The liver failure was more severe,the metabolism of patients with liver failure was more disordered,and the components of exhaled breath in patients with liver failure were more complex,consistent with the patient’s condition. |
