Objective:Establish a stable rat model of chronic prostatitis through the injection of Complete Freund's adjuvant(CFA)into prostate,to investigate the changes of BNP and its receptor NPR-A in spinal cord L6-S1 in chronic prostatitis,And explore the relationship with chronic prostatitis pain.Methods:1.The establishment of the experimental model and the extraction of specimens was finished in the Department of Animal Science,Nanchang University,completed the study of molecular biology in the Urology Institute of the First Affiliated Hospital of Nanchang University.2.Selecting 100 clean type SD male rats,adaptive feed one week,randomly divided into 10 groups,each group include 10 rats: the control group contain 3d,7d,10 d,14d and 28 d subgroups which prostatic injection of 0.1ml physiological saline;The experimental group contain 3d,7d,10 d,14d and 28 d subgroups which Prostatic injection of 0.1ml CFA.3.According to different time points,then extracted the L6 and S1 segments of the spinal dorsal horn from the experimental groups and the corresponding control groups.4.By real-time PCR and Western-blot to detecting the expression of BNP,NPR-A in rats SD L6-S1 spinal dorsal ganglion of experimental groups and control groups.Result:Detecting the expression of BNP,NPR-A in the SD rats spinal cord L6-S1 spinal dorsal ganglion of experimental groups and control groups by real-time PCR and Western-blot,the results show that BNP and NPR-A are overexpression in L6-S1 dorsal ganglion of experimental groups,the 7d and 10 d groups in the rat model of chronic prostatitiswas is more obvious,the difference is statistically significant(p<0.05).Conclusion:By injecting complete adjuvant(CFA)in the rat prostatic can establish stable chronic prostatitis model.The expression of BNP,NPR-A in the spinal cord L6-S1 DRG of the experimental groups rat model of chronic prostatitis is higher than the control groups.BNP ?NPR-A may be involved in the regulation of chronic prostatitis inflammatory signaling pathway,and may act as a suppressor in the signaling pathway.
