| Objective:Establish a stable rat model of chronic nonbacterial prostatitis(CNP)through the injection of Complete Freund's adjuvant(CFA)into prostate,detecting the expression of B-type Natriuretic peptides(BNP)and Protein Kinase G(PKG)in the Spinal cord L6-S1 dorsal ganglion(DRG)of rat chronic nonbacterial prostatitis model and the expression of PKG in the Spinal cord L6-S1 dorsal ganglion of rat chronic nonbacterial prostatitis model after intrathecal injection of BNP in the experimental group and control group.Observing the expression of BNP,PKG and the mutual relationship between BNP and PKG,Preliminary discussing the correlation between BNP,PKG and chronic prostatitis / pelvic pain syndrome(CP / CPPS).Methods: Part one:1.Selecting 80 clean type SD male rats,adaptive feed one week,randomly divided into 8 groups,each group include 10 rats: the control group which Prostatic injection of 0.1ml physiological saline contain 3d,7d,10 d,14d subgroup;The experimental group which Prostatic injection of 0.1ml CFA contain 3d,7d,10 d,14d subgroup.2.Detecting the expression of BNP,PKG in rats SD L6-S1 spinal dorsal ganglion of experimental group and control group by real-time PCR and Westernblot.Part two:1.Selecting 150 clean type SD male rats,adaptive feed one week,randomly divided into prostatic injection of CFA and Intrathecal injection of physiological saline group,prostatic injection of CFA and Intrathecal injection of BNP group,prostatic injection of physiological saline and Intrathecal injection of physiological saline group,each group include 50 rats.A stable model of CNP was established at the 7th day after prostatic injection of CFA and then accept intrathecal injection,each group include 5 subgroup which accept continuous intrathecal injection 1d,2d,3d,4d,5d respectively.2.The BNP group which accept intrathecal injection of BNP,the other groups were intrathecal injection of physiological saline,L6-S1 spinal dorsal ganglion were extracted after continuous intrathecal injection 1d,2d,3d,4d,5d.4.Detecting the expression of PKG in the rats SD L6-S1 spinal dorsal ganglion of each group which after continuous intrathecal injection for 1d,2d,3d,4d,5d.Result:1.Detecting the expression of BNP,PKG in the rats spinal cord L6-S1 spinal dorsal ganglion of experimental group and control group in part one by real-time PCR and Western-blot,the results show that BNP and PKG are overexpression in L6-S1 dorsal ganglion of 3d group,7d group,10 d group which in the rat model of chronic prostatitis,the difference is statistically significant(p <0.05).2.Detecting the expression of PKG in the rat spinal cord L6-S1 spinal dorsal ganglion of prostatic injection of CFA and Intrathecal injection of physiological saline group,prostatic injection of CFA and Intrathecal injection of BNP group,prostatic injection of physiological saline and Intrathecal injection of saline group in part two.We found that the upward of PKG in the rats spinal cord L6-S1 spinal dorsal ganglion of prostatic injection of CFA and Intrathecal injection of BNP group,comparing with other groups,the difference is statistically significant(p <0.05).Conclusion:The overexpression of BNP,PKG in the spinal cord L6-S1 DRG of the rat model of chronic prostatitis,the Intrathecal injection of BNP after successful modeling could upregulate the expression of PKG in the L6-S1 DRG of the rat model of chronic prostatitis.BNP/NPR-A / PKG / BKca signaling pathway is affinity related to chronic prostatitis pain,it may be involved in the occurrence,maintain and regulation of chronic prostatitis pain. |
