Objective:To study the significance of leptin and leptin receptor(OB-R)in pathogenesis of rats with pulmonary arterial hypertension(PAH)induced by monocrotaline(MCT).Methods:15 SD rats were randomly divided into the control group(n=5)and two experimental groups(n=10).The experimental rats were intraperitoneally induced by MCT(60mg/kg),and the control rats with the same dose of 25 percent alcohol.After venous blood was sampled,the mean pulmonary artery pressure(mPAP)of two experimental groups were measured respectively at the second and forth weeks of injection of MCT,and then lung tissue and the heart were removed.The right ventricular(RV)free wall,interventricular septum(S)and left ventricular(LV)were separated and weighted,then right ventricular hypertrophy index(RVHI)was calculated,which was [RV/(LV + S)].Histopathological change of lung tissue was observed with hematoxylin-eosin(HE)and the vessel wall area%(WA%)was calculated to reflect the degree of remodeling of small pulmonary arteries.The expression of serum leptin was detected by ELISA.The expression of OB-R in lung tissue was tested by Western blotting.Results:The lung tissue of control group was pink,smooth,elastic,and with no bleeding point,while the lung tissue of experimental group rats gradually became darker,worse elastic,and with scattered bleeding points over time.As the disease aggravated,WA% increased gradually after the calculation.Compared with the control group,mPAP and RVHI in experimental groups were significantly higher(P<0.05),the expression of leptin in blood serum and OB-R in lung tissue increased significantly(P<0.05),and were positively correlated with mPAP(r= 0.861,r=0.912;P<0.05).Conclusion : 1.Duplicating rat model of pulmonary arterial hypertension induced by monocrotaline was simple and convenient,which the method was worthy of popularizing.2.Leptin and OB-R may play an important role in the pathogenesis of PAH induced by MCT.
