Lung cancer is currently the world’s largest cause of death caused by cancer,the world each year about 150 million people died of lung cancer.In recent years,although lung cancer surgery and radiotherapy and chemotherapy treatment has made some progress,but the cure rate of lung cancer is still disappointing,and 5-year survival rate is also maintained at a relatively low level,about 15%.Lung cancer according to histological type,can be divided into non-small cell lung cancer and small cell lung cancer two.Among them,non-small cell lung cancer accounts for about 85%to 90%of the total number of lung cancer,which is divided into lung squamous cell carcinoma(about 25%to 30%),lung adenocarcinoma(about 40%)and lung Cancer(also known as lung undifferentiated cancer,about 10%to 15%).In general,there was no significant difference in the prognosis of these three subtypes of non-small cell lung cancer.The main cause of poor prognosis in non-small cell lung cancer is distant metastasis,distant metastasis can occur in patients with non-small cell lung cancer early.This explains why non-small cell lung cancer patients will continue to relapse after resection of the primary lesion.It is always a matter of debate about why a tumor can be transferred at an early stage.Comparison of the academic community recognized the view that the tumor on the tumor cells through epithelial-mesenchymal transition from the more stable epithelial tumor cells into mesenchymal tumor cells,the transformation of the tumor cells to make a more aggressive(?)form circulating tumor cells,with the blood flow spread throughout the body to form a tumor metastasis.Epithelial-mesenchymal transition refers to the biological processes of epithelial cells that are transformed into a mesenchymal cell by a specific procedure,and that multiple pathways play an important role in this process.Hippo pathway Hippo was first discovered in the study of Drosophila functional genetic screening,and is a highly conserved signal transduction pathway.In recent years,it has been found that the abnormal state of the hippo pathway and the overactivation of the TAZ gene downstream are thought to have epithelial mesenchymal transition and play an important role in the development of the tumor.Therefore,we intend to detect non-small cell lung cancer patients with different types of circulating tumor cells and the number of TAZ gene expression,combined with clinical data and surgical analysis of patients to assess the disease and evaluation of patients with non-small cell lung cancer Surgical results provide a good basis.This is the first study using CanPatrolTM technology for detection of circulating tumor cells in Guangzhou surexam biotechnology Limited by Share Ltd development,gene localization by circulating tumor cell nucleic acid probe RNA for collection,and the collected cells were classified and identified.In the identification process,innovative to join the sequence of vimentin and twisted protein,which can completely isolate circulating tumor cells in the interstitial cells and double cell phenotype.We can count the different circulating tumor cells and evaluate the patient’s condition with the clinical pathological data.Also,we are on the detection of circulating tumor cells capture and expression of epithelial mesenchymal transition of tumor related gene TAZ,according to the information of TAZ gene positive rate of different clinical pathology,to evaluate the patient’s condition.MethodForty patients with non-small cell lung cancer(NSCLC)who underwent radical resection of thoracic surgery were enrolled in the Southern Medicine Hospital of Southern Medical University.Forty patients with peripheral venous blood were collected on the first day and the fifth day after operation.The typing of circulating tumor cells and the expression of TAZ gene were detected by RNA probe technique.Main results:1.Early non-small cell lung cancer patients with circulating tumor cells,the positive rate.2.The epithelial circulating tumor cells increased gradually with the increase of TNM staging in non-small cell lung cancer.The results were statistically significant(0.5 vs.1.5 vs.5.0,P = 0.037).The positive rate of TAZ gene in circulating tumor cells of EGFR wild type non-small cell lung cancer was significantly higher than that in EGFR mutant non-small cell lung cancer(49.1%vs.29.8%,P= 0.047).The positive rate of TAZ gene was significantly higher in mediastinal lymph nodes(49.1%vs.29.8%,P=0.047),high TNM stage(49.1%vs.29.8%,P = 0.047),CEA abnormality(60.0%vs.22.7%P = 0.014)and NSE(63.8%vs.47.0%,P = 0.042)abnormalities in non-small cell lung cancer patients.5.The positive rate of TAZ gene was decreased before and after radical operation.6.We used tool(http://kmplot.com/)to analyze the survival of patients with TAZ expression and non-small cell lung cancer(Kaplan-Meier).The results showed that higher TAZ expression was associated with poorer overall survival(OS)in patients with non-small cell lung cancer(P<0.001).ConclusionWe detected the number of circulating tumor cells and the expression of TAZ gene in patients with non-small cell lung cancer.It was proved that the number of circulating tumor cells and the positive rate of TAZ gene were different with the clinical characteristics of patients.Monitoring of clinical treatment has a guiding role.
