| Objective:To observe the effect of Danxi gout decoction on hyperuricemia(hyperuricemia,HUA)in rats with metabolism of uric acid and xanthine oxidase(xanthine oxidase,XOD),uric acid(Uricase)enzyme activity,reduce blood uric acid(SUA)and explore its mechanism of action.Methods:40 clean grade SD male rats were randomly divided into 4groups:control group,model group,Danxi gout decoction group,allopurinol group,10 rats in each group.In addition to the blank group,the other groups were treated with yeast paste combined with the mixture of oxygen and potassium chloride to establish the model of hyperuricemia.Continuous intragastric administration of 14 D.Blood uric acid and biochemical indexes of abdominal aorta were measured at 2h after the last one;the XOD and Uricase activity of liver tissue were detected by PBS method,and then was detected by ELISA method.Results:After administration of 14 d,Danxi gout decoction can significantly reduce SUA levels in HUA rats,there was significant difference compared with model group(P<0.05),compared with the control group had no significant difference(P>0.05);Danxi gout decoction can reduce the activity of Uricase increased XOD and the activity of liver tissues,there was significant difference compared with model group(P<0.05),compared with the control group had no significant difference(P>0.05).Conclusion:1,Danxi gout decoction has the effect of anti hyperuricemia,which can significantly reduce the blood uric acid level of model rats;2,The mechanism of Danxi gout decoction on blood uric acid may be achieved by increasing the activity of Uricase in the liver tissue of the rat liver and reducing the XOD activity of the liver tissuel of HUA model;3,Danxi gout decoction has no effect on liver and kidney. |
