The Correlative Research Of MGP And Aortic Valves Calcification

BackgroundThe incidence of calcific aortic valve disease(CAVD)increases with aging,but the mechanism is unclear.CAVD is a complex process of valves differentiation as osteogenesis.The matrix Gla protein MGP is closely related to the occurrence of cification and is an inhibitor of calcification,which are expressed in serum and valve tissues.Study found that MGP was associated with calcification of the animal aortic valve,but the relationship between MGP and human aortic valve was not reported.The purpose of this study was to identify the relationship between the MGP and the calcification of human aortic valve in the normal and calcified valve.ObjectivesTo test the expression of MGP in the calcification of human aortic valve,to reflect the relationship between the MGP and the calcification of the aorta valve.Methods1.Valves were isolated and preserved from those people who had Marfan syndrome or aortic dissection without valves lesions as control group,10cases;at the same time,the valves acquired from patients who need aortic valves replacement surgeon because of simply aortic valves calcification.This valves were divided into two groups for ultrasound before operation or Intraoperative situation,as experimental group.2.Collected the blood from the two groups respectively 30 minutes before operation,each group 10cases;3.Elisa method,Western blotting were used to measure the MGP levels in serum and aortic valve in the two groups.ResultsAortic valve MGP expression compared with the control group,and severe aortic valve calcification group decreased the expression level of MGP(P<0.05);serum MGP protein levels compared with the control group,and severe aortic valve calcification group reduced the level of MGP(P<0.05).ConclusionsAs calcification exacerbation,the MGP in the serum and the valve gradually decreased,especially in the severely calcified group.As aggravate of valve calcification,the MGP was shown to be reduced trend.
