BackgroundMyocardial hypertrophy is an adaptive response of the heart to acute or chronic hemodynamic overload or non hemodynamic factors.It is a common complication of essential hypertension,atherosclerosis,valvular disease,and so on.Myocardial hypertrophy can significantly increase the risk of heart failure and sudden cardiac death.Atorvastatin and spirnonlactone have effects on left ventricular remodeling and the improvement of myocardial function.ObjectsWe detected the effects of combined with atorvastatin and spirnonlactone on the myocardium left ventricular remodeling and myocardial function through the model of myocardial hypertrophy via coarctation of abdominal aorta.The study aims to provide the expermental basis for the clinical treatment of myocardial hypertrophy.MethodsMale SD rats were used to establish the mode of myocardial hypertrophy via coarctation of abdominal aorta.Six weeks after modeling,the structure and function of the heart were monitored by ultrasonic diagnosis.Then they were randomly divided into four groups.Group A: atorvastatin 10mg/kg;Group B: spirolactone 20mg/kg;Group C:atorvastatin 10mg/kg combined with spirolactone 20mg/kg;Group D: NS;Group E:normal control group.The drugs were used by gavage for four weeks.The indicators of ultrasonic cardiogram of rats were detected including posterior wall of left ventricle(LVPW),left ventricular diameter(LVD),interventricular septum(IVS),inner diameter at end-systole(LVS),ejection fraction(LVEF),shortening fraction(LVFS),and changesof hemodynamics indexes including left ventricle systolic pressure(LVSP),left ventricle end diastolic pressure(LVEDP),maximum ascent rate of left ventricle pressure changes(dP/dtmax),maximum descent rate of left ventricle pressure changes(dP/dtmin)and the myocardium energy consumption changes.The differences between groups were compared by SPSS19.0 statistical software.Results1.Compared with the normal control rats,LVPW,IVS,LVS,LVEDP and myocardium energy consumption of the model rats increased significantly,and LVD,LVEF,LVFS,dp/dtmax,dp/tmin/LVSP decreased obviously.There were statistical significance in the results(P<0.05).The above datas suggest the model rats of myocardial hypertrophy were successful.And the myocardial hypertrophy had affected the cardiac function of rats significantly.2.Group A,B and C reduced LVPW,IVS,LVS,LVEDP and myocardium energy consumption,and increased LVEF,LVFS,dp/dtmax,dp/tmin/LVSP,the differences were statistically significant(P<0.05)The difference in the group C was more obviously compared to other two groups.There was no significant difference between the D group before and after administration of drugs(P>0.05).That is,physiological saline had no effect on the cardiac function of rats.The above datas suggested atorvastatin and spironolactone could improve the cardiac function of rats,and the combined application of atorvastatin and spirolactone have better effects on myocardial hypertrophy.Conclusions1.Used alone of atorvastatin or spirolactone can increase the cardiac function of rats,improved ventricular remodeling,and promote the growth and development of rats promote.2.The combined application of atorvastatin and spirolactone has a good synergistic effect on the left ventricular remodeling,cardiac function and myocardial energyconsumption in rats.The effect is more ideal.
