Effects Of Matrine On Cytokines Of Myocardial Tissue In Rats With Isoproterenol-induced Left Ventricular Hypertrophy

Objectives To investigate the inflammatory cytokines in myocardial tissue of matrine on isoproterenol-induced rats chronic pathological left ventricular hypertrophy, and in order to clarify the mutual effect of matrine anti-left ventricular hypertrophy and cytokines.Methods Rats were injected subcutaneously ISO to establish rat model of chronic pathological left ventricular hypertrophy. Seventy-two male rats were randomly assigned to control group(normal saline), ISO treatment group(5mg·kg-1), matrine-alone group(200mg·kg-1), low-dose, medium-dose, and high-dose matrine pretreatment groups(50, 100 and 200 mg·kg-1, respectively). There were 12 rats in each group. The aforementioned medications were administered once daily to 12 rats of each group for 7 successive days. Rats were anesthetized with urethane. Hemodynamic changes were observed using invasive cardiac function determination technology, and pathological morphology changes of the left ventricular tissues were also observed using the hematoxylin-eosin staining method. IGF-1 and TGF-?1 levels in the peripheral serum were determined by ELISA, IGF-1 and TGF-?1 m RNAs expressions in myocardial tissues were determined by real-time fluorescent quantitative RT-PCR. 27 inflammatory factors in myocardial tissues were screened by Ray Bio? Rat Cytokine Array 3 Kit. Among the 27 inflammatory factors, the ICAM-1, MIP-2, CXCL1 and CNTF were determined by ELISA.Results1. Compared with normal group, rats' left ventricular weight and heart rate in ISO group rats were significantly increased(P<0.05), while LV d P/dt, LVSP and LVEDP in ISO group rats were significantly decreased(P<0.01). On the other hand, myocardial fiber structural disorder, interstitial edema and inflammatory cell infiltration in ISO group rats were visible and accompanied with a large area of necrosis. Levels of IGF-1 and TGF-?1 in the peripheral serum were significantly increased(P<0.05), and the m RNA expressions of IGF-1 and TGF-?1 in the myocardial tissues were remarkably increased(P<0.01). Levels of ICAM-1 in myocardial tissues were dramatically increased(P<0.05), and those of MIP-2, CXCL1 and CNTF were significantly decreased(P<0.05).2. Compared with the ISO group, the ISO-induced left ventricular weights in matrine group rats(50, 100 and 200 mg·kg-1) were significantly reduced(p<0.05). Hemodynamics parameters changes and myocardial fibrosis in medium-dose, and high-dose matrine pretreatment groups rats were improved. In addition to the heart rate and hemodynamics parameters weren't changed in low-dose matrine pretreatment groups rats. Levels of IGF-1 and TGF-?1 in the peripheral serum were decreased, and the m RNA expressions of IGF-1 and TGF-?1 in the myocardial tissues were decreased.Matrine significantly decreased the ICAM-1 level and increased those of MIP-2, CXCL1 and CNTF levels in myocardial tissues with ISO-induced left ventricular hypertrophy.Conclusions It is more strong operability and more high model animal survival rate that low-dose continuous subcutaneous injection of ISO is used to establish rat model of left ventricular hypertrophy. IGF-1, TGF?1 and ICAM-1 might play an important role in the process of promoting left ventricular hypertrophy in ISO-induced rats. Matrine has a significant improved effect of ISO-induced rats chronic pathological left ventricular hypertrophy. The current study revealed that matrine was able to regulated upstream factors, such as IGF-1, TGF-?1 and CNTF, downstream factors, such as ICAM-1, MIP-2 and CXCL1 of NF-??signal passway.
