Study On SH-NIC Double-Ethosomes For Tobacco Withdrawal Syndrome

According to domestic and foreign statistics,smoking has caused a great threat to human health,every year more than 500 million people die of smoking and its related diseases,which accounts for 1/3 of the world smokers.Therefore,for medical workers,how to make the existing smokers quit smoking more effectively and as much as possible has become an important issue.The difficulty of smoking cessation is how to effectively overcome the tobacco withdrawal syndrome.The tobacco withdrawal syndrome(TWS)is defined by the World Health Organization(WHO)as an addiction-dependent chronic disease.Currently,internationally recognized methods of treating tobacco withdrawal syndrome are based on nicotine replacement therapy(NRT).According to the formulations prepared by this method include:nicotine gum,nicotine inhalers,nicotine lozenge and nicotine transdermal patches etc.Under this background,this paper has carried on the following research:1.In accordance with the literature,nicotine(NIC),a small molecular substance with a molecular weight of only 162.23,is suitable to make transdermal patches and dissolves well in the inorganic PHase and organic PHase.This research adopts nicotine as the main drug for hydrogel paste.The preparation has the following advantages:reducing the intake of nicotine and the damage caused by gastrointestinal absorption of organs,and patients are more willing to use it.So nicotine transdermal hydrogel paste is the most suitable and promising preparation for the treatment of tobacco withdrawals syndrome.2.After referring to a large number of quit smoking ancient prescriptions and medical history literature,screening out medicine and food dual-purpose to quit smoking of houttuynia cordata for another active ingredient source.In this research,we use Houttuyn Sodium Disulfide as medicinal components,whereas houttuynium easily oxidized and unstable,so we use the same role with the Sodium Houttuyfonate(SH)as a main drug,which effects Co treatment with nicotine.3.Compared with the market to sell the drug release is nicotine Patch,the hydrogel paste has more obvious advantages.However,the market has not yet to see a smoking cessation hydrogel paste.Hydrogel paste prepared in this experiment has high loading,permeability,sticking,moisture resistance,small irritation and repeated exposure etc.;The research development of modified glutinous rice as a natural rubber tackifier,increasing the stability of the preparation;And because of its good moisturizing ability,it can make the preparations contain water more than the highest level,currently up to 63.2%,the cumulative increase of 20%through the amount of drugs,but also more safe and comfortable.4.This study adopted a new drug carrier,Ethosomes(eth.).On the one hand,after the structure of Sodium Houttuyfonate ethosomes encapsulated,the bad smell covered;on the other hand,it can make the drug penetration rate more constant,SH-NIC ethosomes hydrogel paste within 24 h percutaneous release speed is 2%· h-1;In order to improve the effective components of skin penetration,by screening 2%mint oil natural peppermint oil as enhancers,the cumulative penetration amount reached more than 90.59%,which is 1.39 times that of ordinary pressure sensitive adhesive transdermal patch,and peppermint oil has the effect of smoking cessation,can play a certain therapeutic effect,medicine auxiliary one.5.In this research,determined the detection methods of SH and NIC,determined the encapsulation efficiency and drug loading of SH-NIC double-alcohol.After investigation,to determine the injection preparation of SH-NIC double ethosomes,which is the best;with OD value,drug loading and encapsulation efficiency as the evaluation index,through the Central composite design-Response surface methodology optimized process,formulation and quality inspection.The results showed that the indexes of alcoholic hydrogel paste prepared in this research were good,the skin was not stimulated,placed at room temperature for 18 months,there were no significant changes of the index,it is stable and safe.With reference to the database,so far,has not yet seen the related reports of the use of nicotine and houttuyfonate sodium prepared with alcohol-based hydrogel paste.The SH-NIC bimetallic hydrogel patch prepared by this research can control the release of active ingredients,increase the cumulative permeation of drugs,improve drug bioavailability and increase patient compliance.Therefore,this research provides a theoretical basis for the further study of transdermal administration and preparation of tobacco withdrawal syndrome.
