The Expression Of NMDAR1 In The Colorectal Cancer And The Effection Of MK801 On Cancer Cells

Object To explore the expression of the N-Methyl-D-aspartate receptor type R1(NMDAR1)in the Colorectal cancer tissue and cells,and to examine the effect of NMDAR1 antagonists MK801 on colorectal cancer cells in vitro.To Explore the feasibility of NMDAR1 as a independent predictor of early tumor,prognosis and NMDAR1 antagonists as the feasibility of new antitumor drugs.Methods First,Immunocytochemistry was to examine the expression of the NMDAR1 in the Colorectal cancer tissue and cells;Then to explore the effect of MK801 on the Colorectal cancer cells proliferation by means of the MTT assay;And then to analyze the MK801 works in the Colorectal cancer cells apoptosis by annexin V staining;Finally,research the effect of MK801 on the migration of Colorectal cancer cells by Wound healing assay.Results The NMDAR1 was expressed in both Colorectal cancer tissue and cells;,and mianly express in the cytoplasm;MTT assay showed that MK 801 on the Colorectal cancer cells proliferation have inhibitory effect Apoptosis assay manifested that the MK801 induced Colorectal cancer cellsl apopotsis,And Wound healing assay indicate that after MK801-treated,the Colorectal cancer cells migratory ability significantly reduce;Conclusions In the study,we showed the expression of the NMDAR1 in Colorectal cancer tissues and cells.The NMDAR1 antagonists MK801 inhibit the proliferation of the Colorectal cancer cells,induced Colorectal cancer cells early apopotsis.as well as inhibit cells migration.Thus NMDAR1 can serve as a independent predictor of early tumor,prognosis,and NMDAR antagonists as feasible a new antitumor drugs.
