The investigation of the inhibition of baicalein on pseudomonas aeruginosaswimming,swarmmingmotility andtwitching motilityObjective: To investigate the destructive effect of baicalein on pseudomonas aeruginosa swimming motility, swarmming motility and twitching motility.Methods: the experiment were divided into the control group, the different sub-MIC concentrations of baicalein group and clarithromycin group; The MIC of test drugs were measured by the double dilution method;Made different medium, germ-free toothpick vaccinated an overnight single strain in the tablet,the shape of the zone that bacteria was centered was observed, and the zone of motilities were measured to assess the movement of bacteria.Results: The concentric region diameter formed by the swimming motility of Pseudomonas aeruginosa in different concentrations of baicalein groups have no difference with the control group(P > 0.05) ,and the diameters of different concentrations of baicalein groups and control group is bigger than clarithromycin group (P <0.05).The concentric region diameter formed by the swarming motility of Pseudomonas aeruginosa in all the groups. The surrounding of a circular area is formed of radial-like gear in the control group,but smooth can be seen in the baicalein groups with the concentrations of 128?g/ml , 64?g/ml and clarithromycin group. Their diameters of a circular area are smaller than control group (P <0.05). The surrounding of a circular area is sparse and slender in the baicalein group with the concentration of 32?g/ml,but have no difference between the area diameter and control group (P >0.05),the area diameter of different concentrations of baicalein groups is bigger than clarithromycin group; The concentric region diameter formed by the twitching motility of Pseudomonas aeruginosain all the groups. The concentric region diameter both in the different concentrations of baicaleingroup and clarithromycin group are smaller than the control group (P <0.05). The concentric region diameter of clarithromycin group is smaller than the baicalein groups with the concentrations of 32?g/ml (P <0.05),but there are no difference between clarithromycin group and different baicalein groups with the concentrations of 128?g/ml, 64?g/ml (P >0.05).Conclusions: It is not show that Sub-inhibitory concentration of baicalein can inhibit the swmimming motility of Pseudomonas aeruginosa.,but it can inhibit the swarming motility and twitching motility significantly . The baicalein with the concentrations of 128?g/ml and 64?g/ml can inhibit the twitching motility of Pseudomonas aeruginosa and have no difference with clarithromycin.Part ? In Vitro Activity of baicalein in Combination with Cefoperazone/sulbactam on Pseudomonas Aeruginose BiofilmObjective To observe the destructive effect of baicalein on the biofilm of Pseudomonas Aeruginose in vitro and the synergistic antibacterial activity in combination with Cefoperazone /sulbactam.Methods PAO1 was for biofilm formation in vitro. The minimum inhibitory concentrations ( MIC ) of baicalein and Cefoperazone/ sulbactam , and Minimum bactericidal concentration(MBC) of Cefoperazone/ sulbactam were measured by doubling dilution; viable bacterial counts were determined by serial dilution;the configuration of the BF on the carries surface was observed by confocal laser scanning microscope.Result When the biofilm being reacted with drugs for 24 hours, the viable bacterial counts showed that the number of bacteria in biofilm of the baicalein group,Cefoperazone /sulbactam, baicalein plus Cefoperazone/sulbactam were significantly less than that in the control group ( P <0.01);baicalein group plus Cefoperazone /sulbactam was significantly less than that in the baicalein group and Cefoperazone /sulbactam ( P < 0. 01).While there was no difference between baicalein group and Cefoperazone/Sulbactam (P>0.05 ) . The BF formed in the baicalein group using confocal laser scanning microscope to observe which was almost viable cells (green fluorescence) was significantly less than those in the control group,the same as the Cefoperazone/sulbactam,which was the green fluorescence more than the red one.While baicalein plus Cefoperazone/sulbactam were almost dead,which was almost the red fluorescence.Conclusions: baicalein can destroy the formed Pseudomonas Aeruginose BF and significantly destroy the formed the bacterial colony count ofPseudomonas Aeruginose in BF when it is used in combination withCefop-erazone/sulbactam.
