Objective:To investigate the expression of DDX39 and the correlation between DDX39 rs 12461754 polymorphism and the family clustering genetic susceptibility to Hepato cellular carcinoma HCC.Methods:Immunohistochemistry detected the DDX39protein expression.Genotype and alleles of rs 12461754 were deter-mined in 20 HCC family groups(77cases)and 10 normal control family groups(35 cases)in Fusui County through Time of Flight Mass Spectrometer.Results:The DDX39 protein expression in HCC(9.3±0.86307)was higher than in hepatocellular paracancerous tissues(6.28±0.85809)(P<0.05).For the alleles of DDX39 rs12461754 site,the risk of hepatocellular carcinoma(HCC)for indi-viduals with[A]allele was 0.407-fold(95%CI =0.125-1.326,P=0.136)compared with that for individuals with[G]allele among the members of the normal control group,the risk of hepatocellular carcinoma(HCC)for individuals with[A]allele was 0.981-fold(95%CI =0.298-3.326,P=0.976)compared with that for individuals with[G]allele among the members of the non-patients of HCC families.The frequencies of the[A]and[G]alleles were similar in the patients of HCC families and the non-patients of HCC families(P>0.05).For the genotype of the DDX39 rs 12461754 site,the differences in frequencies of AA,AG,and GG genotypes were not statistically significant among the patients of HCC families and the non-patients of HCC families and normal control families.Conclusion:The allele of DDX39 rs12461754 site is not correlated with the occurrence of HCC family clustering in Guangxi.DDX39 protein.is overexpression on HCC patients tissues.
