Effects Of Chloral Hydrate On Cognitive Function The Expression Of Bcl-2 And Bax Proteins In Hippocampus In Rats

?Objective?To investigate the effects of chloral hydrate on cognitive function of bcl-2 and bax proteins in hippocampus in rats.?Metheds?24 male Sprague Dawley rats were randomly assigned 3groups as normal control group,chloral hydrate 3 days group and chloral hydrate 6 days group,8 animals per group.10% chloral hydrate was injected intraperitoneally at the dose of 400mg/kg in the chloral hydrate 3 days group for three days,the chloral hydrate 6 days group rats for six days,and the normal control group was injected with same amount of saline for six days.After the above processing,the ability of learning and memory was evaluated by the Morris water maze test,The expression of bcl-2?bax proteins in hippocampus were detected by immunochemistry.The structure changes of Neuroal morphology were observed by HE stain.?Results?(1)Morris Water Maze test : There were no statistical differences in the escape latency on the 5th day and the target crossings on the 6th day among the 3 groups(P>0.05).Results of escape latency are as followed:control group(10.107±4.761s),3d CH group(8.949±3.175s),6d CH group(10.673±4.327s).Results of target crossings are as followed : normalcontrol group(2.5±0.949),3d CH group(3.313±1.438),6d CH group(3.000±1.035).(2)immunochemistry : Compared to the normal control group,the expression of the Bcl-2 and Bax protein in the 3d CH group and 6d CH group didn't make significant difference(P>0.05).Results of Bcl-2 protein expression are as followed : AOD was 0.233±0.057 for control group,0.226±0.034 for 3d CH group,0.245±0.025 for 6d CH group.Results of Bax protein expression are as followed:AOD was 0.210±0.017 for control group,0.222±0.033 for 3d CH group,0.199±0.029 for 6d CH group.The ratio of Bcl-2/Bax are as followed : control group(1.120±0.306),3d CH group(1.036±0.200),6d CH group(1.224±0.179).(3)HE staining:The neurons in the CA1 area of hippocampus lined up tightly and formed round or elliptical with clear nucleolus,nerve cells atypia was not obvious.?Conclusion? While small doses of chloral hydrate was used repeatedly for many times in the short term,it has no obvious effect on cognitive function in rats,the expression of bcl-2 and bax in hippocampus didn't make significant difference.
