Objective: The aim of the study was to investigate the relationship between the APOC3 rs5128 polymorphism and plasma lipid levels and the severity of coronary stenosis in the coronary heart disease(CHD)patients with different body mass index(BMI)in Chinese Han population.Methods: The Three hundred and twelve inpatients who were confirmed with coronary heart disease by coronary angiography examination at the Department of Cardiology,the Affiliated Hospital of North Sichuan Medical College(Nanchong,China)were enrolled in the study from April 2014 to July 2015.Three hundred and twelve CHD patients were divided into healthy weight group [205 patients,BMI =(22.30 ± 1.88)kg/m2] and overweight/obese group [107 patients,BMI =(27.64 ± 2.48)kg/m2].According to the TG value,the patients were divided into normal TG group[219 patients,TG =(1.09 ± 0.31)mmol / L] and high TG group [93 patients,TG =(2.78 ± 1.66)mmol / L].Physiological,biochemical and coronary angiographical data were collected for all patients.The severity of coronary stenosis was assessed by the Gensini scoring system.Genome DNA was extracted from peripheral blood leucocytes and the genotypes of the APOC3rs5128 polymorphism were determined by polymerase chain reaction-restriction fragment length polymorphism method.Results: The overweight/obese patients had higher weight,BMI,prevalence of hypertension,triglyceride(TG),lipoprotein(a),TG/high-density lipoprotein cholesterol(HDL-C),total cholesterol(TC)/HDL-C,low-density lipoprotein cholesterol(LDL-C)/HDL-C and apolipoprotein B100(APOB100)/apolipoprotein AI(APOAI),and lower HDL-C and APOAI than the patients with healthy weight(P<0.05 for all).The results of comparison between the normal TG group and the high TG group showed that TC,LDL-C,VLDL-C,APOB100,TG / HDL-C,TC / HDL-C,LDL-C / HDL-C and APOB100 / APOAI levels were higher than those in normal TG group,HDL-C level was lower than normal TG group(P<0.05 for all).In healthy weight group,the G carriers had higher prevalence of hypertension than the patients with the CC genotype(P<0.05).In overweight/obese group,the G carriers had higher levels of TG and TG/HDL-C than the patients with the CC genotype(P<0.05 for both).In healthy weight group and overweight/obese group,there were no significant differences in the distributions of the genotypes or alleles of the rs5128 polymorphism among the tertiles of the Gensini scores.There was no significant difference in the distribution of the APOC3 rs5128 polymorphism allele frequency in the numbers of diseased coronary vessels.There was no significant difference in coronary Gensini score between normal TG group and high TG group.And there were no significant differences no matter between the Gensini score and the TG level in the subgroup of different stenosis,or between the TG level in different stenosis subgroups(P<0.05 for both).No matter in healthy weight group or in overweight and obese group,the TG levels in the different coronary stenosis subgroups were no significant differences.There were no significant differences in the distribution of APOC3 rs5128 polymorphism between the Lipid [LP(a),LDL-C,TG,TC,HDL-C,APOB100]normal groups and theLipid elevated / decreased groups(P<0.05 for both).Conclusion: The APOC3 rs5128 polymorphism is significantly associated with plasma TG and TG/HDL-C levels,but not with the severity of CHD.There are racial differences in the APOC3 rs5128 polymorphisms and its correlation to atherosclerosis and CHD.
