Objective To study the difference of Nerve growth factor receptor expression in the Nestin positive or negative cholinergic neurons in basal forebrain of healthy adult SD rats,explore the possible mechanism of the difference of neuronal function.Method Forty adult rats weight about 200-250 g were selected,without gender,using paraformaldehyde perfusion to stabilized the model to get rats' brain slices,the rats' brain slices were divided into single-standard group ? double standard group and control group.single-standard groups: Nestin,ChAT,TrkA,double immunofluorescence labeling of the Nestin and TrkA,ChAT and TrkA.The total expression of three neurons in the basal forebrain was observed by immunofluorescence double staining methods.The distribution of the Nestin,ChAT and TrkA in the basal forebrain,the vertical branch of the oblique band and the number of the relative positive reactant were detected by immunohistochemistry and confocal microscopy methods.Counting the number of Nestin,TrkA and ChAT immunoreactive cholinergic neurons in the same area of basal forebrain(MS,v DB,h DB)and also count the number of the ChAT immunoreactive neurons express TrkA,the Nestin positive neurons express the TrkA.To speculate the Nestin negative neurons express the TrkA.We used a statistical mean ± standard deviation(`X ± S)to analyze the data.Results The results showed that the TrkA positive cells were found in the MS-DBB region of the basal forebrain of rats.The TrkA,Nestin cholinergic neurons and ChAT have the co-localization in the same area,Three neurons were distributed in the MS-DBB region of the basal forebrain of rats.The data shows that the number of the ChAT positive neurons is higher than Nestin positive neurons,All Nestin-ir neurons showed ChAT immuoreactivity;Approximately 90% of Nestin immunoreactive neurons express TrkA,about 70% of ChAT-immunoreactive(ir)neurons showed TrkA immunoreactivity,Nestin negative neurons express TrkA.Conclusion The Nestin neurons can express the TrkA,but the number of two kinds Nestin neurons is different.The number of the TrkA expressed higher in the Nestin positive neurons than Nestin negative neurons.It was confirmed that Nestin positive neurons had combined more nerve growth factor.The Nestin positive or Nestin negative cholinergic neurons in terms have significant difference in plasticity,excitability,Anti-injury function and the other aspects,and the difference may be due to the expression number of different nerve growth factor receptors.
