| Objective: Neuroglobin(NGB) , as a new globin, has endogenous protective function.By discussing the expression of neuroglobin in brain of rat with infectious shock and the relevance of neuroglobin and ROS、caspase-3、PI3-K/AKt, to study the protective effect and mechanism of neuroglobin.Methods: Sixty SD rat were randomly divided into two groups of 30 rat each:Control group(group C) and LPS group(group M). The rats in group M were injected LPS 10mg/kg through tail vein to built infectious shock model,and the control group were given equal volume NS. Monitor hemodynamics, the success standard of infectious shock is systolic blood pressure dropping to 60% of the basic values. The levels of rat TNF-α and IL-6 in serum at the time points of before LPS injection (T0)、1h(T1)、2h(T2)、4h(T3)、6h(T4)after LPS injection were measured using ELISA Kits.ROS was studied by flow cytometry. NGB、caspase-3、CHOP and PI3-K/AKt protein expression in brain were determined using Western blotting.Result: Compared with group C and T0, the TNF-α、IL-6 concentrations in serum and caspase-3 in brain were increased significantly at T1~T4(P<0.05) ,NGB protein content in serum and brain at T3、T4 were increased significantly(P<0.05); The expression of CHOP、ROS and PI3-K/AKt in brain was increased significantly at T4 compare to group C (P<0.05). Mitochondria in slight swelling and endoplasmic reticulum obviously disintegrate.Conclusion: Apoptosis and free radical damage during infectious shock. NGB was over-expressed in brain of rats with infectious shock, which may play a role of protective function through endogenous anti-apoptotic approaches and PI3-K/AKt anti-apoptotic pathway. |
