Objective: Determination of primary glioma cells in vitro sensitivity to different concentration of doxorubicin by MTT method.Which provide the perspective of basic experiment to research glioma primary cells sensitivity to different concentration of doxorubicin in vitro.Methods: Based on 27 cases of brain glioma patients(postoperative pathological biopsy are confirmed for glioma)tumor tissue in resection to make primary cell culture,which divided into three groups according to the WHO classification.Determination of primary glioma cells in vitro sensitivity to different concentration of doxorubicin by MTT method.Results: Under the condition of in vitro culture.With the increase of the concentration of doxorubicin and the extension of time,the inhibition rate of primary glioma cells is on the rise.Under the action of the different concentration of doxorubicin,the sensitive rates between WHO grade ? primary glioma cells has significant difference(P<0.001),the sensitive rates between WHO grade ? primary glioma cells has significant difference(P<0.001),the sensitive rates between WHO grade ? primary glioma cells has no statistical difference(P>0.05).For the whole group of 27 cases of samples,the sensitive rates between primary glioma cells has significant difference under the action of the different concentration of doxorubicin(P<0.001).Conclusions: Primary cells glioma can be extracted effectively by enzyme digestion method.Under the condition of in vitro culture.Doxorubicin has inhibitory effect to the growth of primary glioma cells.With the increase of the concentration of doxorubicin and the extension of time,the inhibition rate of glioma cells is on the rise.There are large differences inhibition rate between different individuals.The higher level of tumor,the bigger the difference.There are differences sensitivity in different grade primary glioma cells to doxorubicin.
