Objective: Through the rabbit ear to create a hypertrophic scar model,scar injection of ginsenoside Rb1,to observe the therapeutic effect and explore the relevant mechanism.Methods: 8 New Zealand rabbits were selected randomly and 6 round wounds were made in each ear to establish the model of hypertrophic scars.After the wounds epithelialization,the hypertrophic scars were separated into 4 groups: Blank control group(N),Dexamethasone control group(D),Sodium chloride control group(S),Ginsenoside Rb1 treatment group(R).The concentration of D and G was 1mg / ml.The drugs were injected into the scars once every 3 days for three times.On 1st,2nd,4th,8th weeks after the treatment,2 rabbits’ s samples(24 cases)were chosen to test HE staining,Masson staining and detect PCNA expression by immunohistochemistry,and we determined the expression of m RNA of transforming growth factor beta1 and Collagen I by reverse transcription polymerase chain reaction(PCR).The data for each group was divided by the blank group data for data normalization.The measurement data with " x ±s" form were processed by software SPSS19.0 for variance analysis,if the data didn’t meet the normality and variance homogeneity,switch to nonparametric test.Electionα= 0.05,when P ≤ 0.05,the data was statistically significant.Result: There was no statistically significant difference between the three groups on the first and second weeks after administration.On 4 and 8 weeks after injection,HS scar improved significantly in dexamethasone and ginsenoside Rb1 group than in the saline group,Scar elevation index decreased(P <0.05),the expression of PCNA was decreased(P <0.05),the expression of TGF-β1 and typeⅠcollagen m RNA decreased(P <0.05).But there was no statistically significant difference between dexamethasone and ginsenoside Rb1 group.Conclusion: Local injection of ginsenoside Rb1 can inhibit the proliferation of active cells,reduce angiogenesis,inhibit the expression of TGF-β1,reduce collagen synthesis,effectively improve the proliferation of rabbit ear scar.This finding may be provide theoretical basis for the application of ginsenoside Rb1 on human hypertrophic scars.
