Objective: To surch the Corydalis saxicola bunting extractive inhibit F-actin Polymerization in A549 cells and its possible mechanism.Methods : MTT assay was used to detect the proliferation of corydalis saxicola bunting extractive in different concentrations(1,2.5,5,10,20mg/L)respectively effect 48 h,72h.Wound healing and transwell was used to detect the migration of in different concentrations after drug-treated.A549 cells after different concentrations(1,2.5,5mg/L)drug-treated 48 h were immunostained with anti-paxillin antibody and rhpdamine phalloidin to observe focal adhesions and F-actin stress fibres.Through Westernblot,we detected the different expression of paxillin,Y31-paxillin,cofilin-1,S3-cofilin-1.Through si RNA knockdown Cdc42 in A549 cells,we detected the different of migration and the different expression of MAPKs.Results: The MTT assay showed corydalis saxicola bunting extractive can reduce A549 cells proliferation in concentrations(20mg/L)after drug-treated 48 h and in concentrations(10,20mg/L)after drug-treated 72h(P<0.05).The wound healing and transwell assay showed corydalis saxicola bunting extractive2.5,5mg/L reduce the migration rate of A549 cells(P<0.05).A549 cells was interfered with cytoskeletal remodeling in vitro.To compare the control,the express of S3-cofilin-1was downregulation on 5mg/L group,but total cofilin-1,paxillin and Y31-paxillin have no statistical difference.Cdc42 knockdown A549 cells induced a reduction in migration.However,the expression of MAPKs have no statistical difference.Conclusions:Corydalis saxicola bunting extractive reduce the migration rate of A549 cells,increase actin filament disassembly,increase the activity of cofilin-1;.Cdc42 knockdown A549 cells induced a reduction in migration.
